复方银黄微型灌肠剂对巨噬细胞分泌肿瘤坏死因子和白细胞介素-6的影响

摘　　要：目的研究复方银黄微型灌肠剂(Yinhuang Micro-enema Compound,YHMEC)含药血清对大鼠腹腔巨噬细胞(PMФ)分泌肿瘤坏死因子(TNF-α)和白介素-6(IL-6)的影响。方法将大鼠随机分成3组,分别于末次灌肠给药后10,30,60 min采集含药血清,采用MTT比色法检测不同采血时间的含药血清对PMФ活力的影响;细胞培养上清液中TNF-α,IL-6的表达采用ELISA法检测。结果末次给药后30,60 min采集的含药血清对PMФ的活力及细胞因子的表达影响显著(P<0.05或P<0.01)。结论复方银黄微型灌肠剂含药血清能显著抑制LPS诱导的TNF-α,IL-6表达,具有明显的体外抗炎活性。Objective To investigate the effects of Yinhuang Micro - enema drug - containing serum on the secretion of TNF-α and IL- 6 in mouse（PMФ）. Methods The mice were randomly divided into three groups, and their serum was collected after rectal administration of 10,30,60 min, the effects of drug - containing serum which were collected at different time on PMФ） viability were determined by MTT method, and the levels of TNF-α and IL - 6 in the medium were measured by ELISA method. Results The serum which was collected after rectal administration of 10,30,60 min had significant effect on the PMФ viability and secretion of TNF-α, IL - 6 （ P 〈 0.05 or P 〈 0. 01 ）. Conclusion YHMEC drug - containing serum can significantly suppress the TNF-α and IL - 6 secretion induced by LPS and has obvious anti - inflammatory effect in vitro.
