紫苏叶提取物抗家兔动脉粥样硬化的实验研究

摘　　要：目的探讨紫苏叶提取物(FPE)对由高脂高胆固醇引起家兔动脉粥样化的影响。方法采用高脂高胆固醇饲喂法建立家兔动脉粥样硬化模型,分别测定正常对照组、高脂模型组、紫苏叶提取物低剂量组(0.17 g.kg-1)、中剂量组(0.5g.kg-1)、高剂量组(1.5 g.kg-1)血清中TC,TG,LDL-C,HDL-C,MDA含量及SOD的活性,大体染色计算脂肪斑块面积比(PA),组织切片观察主动脉内膜增生(IH)。结果FPE组均能明显降低血清中TC,TG,LDL-C,MDA的含量(P<0.01),并能升高血清中HDL-C的含量(P<0.01)和SOD的活性(P<0.05),FPE干预组PA值下降明显,FPE中剂量组、高剂量组与高脂模型组比较具有统计意义[分别为(21.52±7.58)%,P<0.05;(15.82±3.64)%,P<0.01;(53.59±29.31)%]。病理切片显示,各用药组主动脉IH程度均小于高脂模型组,其中高剂量用药组AS程度最小。结论紫苏叶提取物具有抗高脂饮食诱导家兔动脉粥样硬化的作用,其作用机制可能与其调节血脂与抗脂质过氧化有关。Objective To observe the preventive effects of Folium Perillae extract （ FPE ） on atherosclerosis （AS） formation in rabbits. Methods High fat and cholesterol diet was administered to make the AS model of rabbits. The rabbits were divided into 5 groups randomly： NC（normal control）group, HLM （high lipid model） group, LFPE, MFPE and HFPE group （low, middle, high dosage of FPE, which represented 0.17 g · kg^-1 , 0.5 g · kg^-1, 1.5 g · kg^-1 respectively）. The contents ofTC, TG, LDL -C, HDL- C, MDA and the SOD activity in serum were detected; Percentage of plaques covering aortic intima（PA） was calculated by gross stain, aorta intimal hyperplasia （IH） was observed by histological section. Results The three FPE groups could decrease the contents of TC, TG, LDL - C, MDA significantly （ P 〈 0. 01 ） as compared with the HLM group, and the content of HDL - C in HFPE group was increased significantly （ P 〈 0. 01 ）, the same was the SOD. PA was （ 21.52 ± 7.58 ） % （ P 〈 0. 05 ）, （ 15.82 ± 3.64） % （P 〈 0. 01 ）, and （53.59 ± 29.31 ） % in MFPE, HFPE and LFPE groups, respectively. Degrees of IH formation in aorta of all FPE groups were less than that in HLM group. Conclusion FPE can prevent Atherosclerosis formation by high lipid and cholesterol feeding. The mechanism maybe concern with blood -fat adjustment and anti -liopid peroxidation.
