摘 要:目的探讨由黄连、黄芪和地黄水提物组成的组合物(简称三黄组合物,SH)对2型糖尿病(DM)大鼠糖脂代谢的影响并初探其机制。方法以高糖高脂饲料喂养SD大鼠两个月后小剂量腹腔注射链脲佐菌素(STZ)(25 mg.kg-1),连续注射3 d,建立糖尿病大鼠模型,随机分为模型组(MC),盐酸二甲双胍组(M et),三黄组合物低剂量组(SHL)、中剂量组(SHM)、高剂量组(SHH),外加常规饲料喂养的正常组。给药后第3,10周测空腹血糖水平(FPG),第7周测糖异生水平。于实验结束时腹主动脉取血,测血清胰岛素(Insu lin),总胆固醇(TC),甘油三酯(TG),高密度脂蛋白胆固醇(HDL-C),低密度脂蛋白胆固醇(LDL-C),谷丙转氨酶(ALT),尿酸(Uric ac id),游离脂肪酸(FFA)含量,取肝脏测肝糖原含量。结果与模型组相比,SH各剂量组都能显著降低糖尿病大鼠的血糖水平,以中剂量组效果最明显,与二甲双胍组相近;三黄组合物可以降低DM大鼠TC,TG,LDL-C含量,升高HDL-C水平,促进胰岛素分泌和肝糖原的合成,抑制糖异生,调节血清FFA,ALT的含量。结论三黄组合物可以有效降低糖尿病大鼠的血糖水平,调节血脂异常,其机理可能包括保护和修复胰岛,促进胰岛素分泌,抑制糖异生,促进肝糖原合成等,此外,三黄组合物对大鼠肝脏也有一定的保护作用,对抑制尿酸沉积,提高尿酸清除率,从而改善胰岛素抵抗,保护肾脏,防治DM并发症具有积极作用。Objective To observe the effects of SanHuang ( Rhizoma Coptidis ; Radix Astragali; Radix Rehmanniae, SH ) compound on metabolism of glucose and lipid in Type Ⅱ diabetes mellitus(DM) rats. Methods SD rats were fed with high glucose and high fat diet for 2 months, then given ip STZ (25 mg · kg^-1 ) for successive 3 days to establish diabetes mellitus (DM) rat model, into 5 groups randomly : model control group ( MC), metformine hydrochloric acid group ( Met), SH low dose (SHL) , SH middle dose ( SHM), SH high dose (SHH) groups, and normal group which was fed with normal diet. After i.g. medication for 3 and 10 weeks, respectively fasting plasma glucose( FPG), and gluconeogenesis on the 7th week were detected. At the end of experiment, the contents of insulin, total cholesterol (TC), triglyceride (TG), high- density lipoprotein cholesterol (HDL-C), low - density lipoprotein cholesterol ( LDL - C), ALT, uric acid, free fatty acids (FFA) and hepatic glycogen were determined. Results Compared with model group SH different dose group could reduce plasma glucose level of DM rats obviously, especially SH M group was the most obvious and close to Met. SH could reduce the levels of TC, TG, LDL - C, FFA, ALT, uric acid, raise HDL - C, enhance insulin secretion and glycogenesis and inhibit gluconeogenesis. Conclusion SH can reduce plasma glucose level of DM rats efficiously, regulate hyperlipidemia and the mechanism may include protecting and repairing pancreas, enhancing insulin secretion and glycogenesis, inhibiting gluconeogenesis and so on. In addition, SH can also protect liver, has positive effect on ameliorating insulin resistance( IR), protecting kidney and preventing and curing DM complication through inhibiting deposition and raising clearance rate of uric acid.
