摘 要:目的研究柴胡皂苷-d(SS-d)对乙醇损伤大鼠原代培养肝细胞保护的作用和机制。方法采用胰蛋白酶消化、分离大鼠肝细胞进行原代培养,乙醇体外诱导肝细胞损伤,以不同浓度的SS-d对肝细胞保护,检测培养液中丙氨酸氨基转移酶(ALT)活性和肝细胞内丙二醛(MDA)含量、谷胱甘肽过氧化物酶(GSH-PX)的活性,MTT法检测肝细胞存活率。结果SS-d(1.0,2.0,3.0mg.L-1)明显改善肝细胞存活率,抑制乙醇引起的ALT活性的升高,对肝细胞中MDA含量升高和GSH-px活性降低均有明显的抑制作用。结论SS-d对乙醇损伤大鼠肝细胞有保护作用,其机制可能与SS-d清除自由基、抑制质脂过氧化作用有关。Objective To study the protective effect and mechanisms of saikosaponin -D (SS -d) on primary rat hepatocytes injured by ethanol. Methods Primary hepatocytes were isolated,cultured by trypsin digestion and induced by ethanol. Various concentrations of SS - d were added to the primary cultured hepatocytes for 12h. The content of malondialdehyde(MDA) and activity of glutathion peroxidase( GSH- px) in hepatocytes and the activity of ALT in cultural supernatant were determined by general methods. The viability of hepatocytes was measured by MTT. Results Compared with model group, SS - d could restored remarkably ALT level in supermatant of cultural hepatocytes, decreased MDA content, elevate GSH - px activity of hepatocytes , and improve hepatocyte viability. Conclusion SS - d has protective action on primary rat hepatocytes injured by ethanol. The mechanisms may be related with eliminating free radicals and reducing lipid peroxidation.
