摘  要:利用原代犊牛睾丸细胞从临床上表现口疮症状的羔羊痂皮材料中分离获得1株病毒,对该株病毒进行病毒形态学、理化学、PCR检测与动物回归试验等系统鉴定后,证实该分离株为羊传染性脓疱病毒(Orf virus,ORFV),命名为ORFV-sy。利用PCR方法克隆出其B2L基因,并将该基因序列和推导的氨基酸序列与6个不同来源的ORFV毒株进行同源性和亲缘关系的比较分析。结果表明,各毒株间核苷酸和氨基酸的同源性分别为97.8%~99.5%和97.6%~99.5%;系统发育进化树结果表明,ORFV-sy毒株与印度分离绵羊株ORFV-Mukteswar67/04、ORFV-Izatnagar79/04的亲缘关系较近,表明不同地区分离毒株的B2L基因差异不大。An Orf virus strain(ORFV-sy) was successfully isolated from crust materials of lambs with clinical sore mouth symptom from a sheep farm of Songyuan city of Jilin province by passaging in calf testis cells. The virus was identified as Orf virus based on morphology, physico-chemical property, PCR results and animal regression test. The B2L gene of ORFV-sy strain was amplified by PCR which shared 97.8%-99.5% homology of nucleotide sequences and 97.6%-99.5% homology of amino acids with those of 60RFV strains. The results of the phylogenetic tree analysis demonstrated that there was a close genetic relationship between ORFV-sy strain and ORFV-Mukteswar 67/04,0RFV-Izatnagar 79/04 isolated from India. The result indicated the differences of different strains was not obvious.
