摘 要:采用RT-PCR和免疫荧光染色技术,分别检测T668基因mRNA及蛋白在旋毛虫不同发育时期中的时空表达情况。RT-PCR结果显示,该基因从旋毛虫新生幼虫出生即开始转录,随着日龄的增加,T668基因mRNA逐渐减少,在感染后11~14 d T668基因的转录趋于停止。免疫荧光染色结果表明,在感染后1d,旋毛虫虫体即有T668蛋白表达;感染后6~13d,T668蛋白分泌到肿大的肌细胞核上;14~20d,肿大的肌细胞核上基本观察不到T668蛋白,但整个虫体仍有很强的蛋白表达。这提示在旋毛虫的寄生过程中,T668基因的转录及表达呈一定的时空特点,这可能对旋毛虫包囊形成有着重要作用。To explore the transcription and expression of Trichinella spiralis T668 gene during invasion and nurse cell formation, RT-PCR and immunofluorescence were used to analyze the temporal and spatial expression patterns of T668 in both mRNA and protein. RT-PCR analysis showed that transcription of the T668 gene appeared and maintained a considerable high level at the first day when the newborn larvae was born. With the increase of age,the level of T668 mRNA reduced. From 11 to 14 days postinfection (d. p. i. ) ,there was barely detectable transcription. Immunofluorescence analysis indicated that in the early stages of infection (from 1 to 5 d. p. i. ), the increased expression of the T668 protein was limited to the newborn larvae. At a later stages of infection (from 6 to 13 d. p. i. ), the fluorescence spread to the Trichinella and enlarged nuclei. From 14 to 20 d. p. i. ,the staining was limited to the Trichinella and barely detectable in enlarged nuclei. It is concluded during the parasitism processes of newborn larvae,the transcription and expression of T668 gene is temporal and spatial. These findings provide evidence that the T668 protein could play an important role in nurse cell formation.
