摘  要:探讨了渗稳剂种类、酶种类和浓度、酶解时间、酶解温度、菌龄等因素对黄伞原生质体制备和再生的影响.结果表明,其原生质体制备和再生的最佳条件是菌龄为7d,2%溶壁酶处理2.5h,酶解温度25℃,0.6mol/LKCl为制备时的渗稳剂及0.6mol/L甘露醇为再生时的渗稳剂,其原生质体的产率为2.16×107个/mL,再生率为0.52%.The effects of some factors, such as different osmotic pressure stabilizers, different enzymes, the density of enzyme, the digesting time, the digesting temperature, the age of mycelium of the separation and regeneration of Pholiota adipose protoplasts were studied. The results showed that the highest separation rate of protoplast with 2.16 × 10^7/mL and reproduction rate of protoplast with 0.51% could be obtained under the conditions of 7 days mycelium, 2% lywallzyme enzyme digesting at 25 ℃ for 2.5 hours, applying 0.6 mol/L KCl for osmotic pressure stabilizer of separation and 0.6 mol/L mannitol for osmotic pressure stabilizer of regeneration.
