摘 要:目的观察石菖蒲有效成分β-细辛醚对缺血再灌注损伤(MIRI)心肌细胞线粒体膜电位(MMP)的影响。方法采用连二亚硫酸钠(Na2S2O4)建立心肌细胞MIRI模型,将原代培养的SD大鼠乳鼠心肌细胞分6组:正常对照组、MIRI模型组、β-细辛醚高、中、低剂量组(25,12.5,6.25μg/mL)、基质组。利用流式细胞术(FCM)检测MMP平均荧光强度。结果MIRI模型组心肌细胞MMP明显降低,与正常对照组比较有统计学意义(P<0.05);β-细辛醚能不同程度升高MMP,与模型组比较有统计学意义(P<0.05)。结论β-细辛醚能有效抑制MIRI心肌细胞线粒体的损伤,稳定MMP。Objective To study the effects of β- asarone on mitochondrial membrane potential(MMP) of cardiac myocytes with ischemia reperfusion injury. Methods The cardiac myocytes model of myocardial isehemia reperfusion injury (MIRI) was induced with sodium dithionite (Na2S2O4). The primary cultured cardiac myocytes of SD neonatal rats were randomized into six groups: normal group, model control group, high -dose β -asarone (25μg/mL), middle -dose β-asarone group(12.5 μg/mL), low-dose β-asarone group (6.25 μg/mL) and medium group. Mitochondrial membrane potential(MMP) of myocardial cells were measured by flow cytometry. Results Compared with the normal group, MMP in the model control group was obviously decreased ( P 〈 0.05) ; Compared with model control group, MMP in β - asarone groups was obviously increased ( P 〈 0.05). Conclusion β - asarone can inhibit the injury of myocardial mitochondria effectively, and stabilize MMP.
