摘 要:目的观察肠激安方对肠易激综合征(IBS)模型大鼠肥大细胞的影响。方法对出生后8~21d的大鼠连续性给予结肠内炎症刺激,建立腹泻型IBS的动物模型。采用甲苯胺蓝改良染色法,以光镜和电镜观察肥大细胞数量及其细胞脱颗粒情况。结果肠激安方能提高抬腹和拱背的压力阈值,与模型组相比有显著性差异(P<0.01);光镜及电镜观察显示,肠激安方中、高剂量组肥大细胞计数及脱颗粒百分比显著降低,与模型组相比有显著性差异(P<0.05)。结论肠激安方能降低IBS模型大鼠的肠道敏感性,对肥大细胞脱颗粒有显著改善作用。Objective To observe the effect of Changji' an Prescription (CP) on depressing the mast cells in the irritable bowel syndrome(IBS)rats. Methods The postnatal rats aged 8 -21 days were given continous inflammation stimulation inside the colon to replicate the animal model of the IBS. The number of mast cells and their degranulation were observed by modified Toluidine Blue staining method under light microscope and electron microscope. Results The pressure threshold value of rats raising their abdomen and back in the model group was obviously lower than that in the normal group ( P 〈 0.01 ). After medication for two weeks, the pressure threshold value of rats raising abdomen and back was obviously higher than that in the model group ( P 〈 0. 01 ). Observation results under light microscope and electron microscope showed that the number of mast cells and the percentage of mast cells with degranulation in the model group were obviously higher than those in the normal group ( P 〈 0. 05 ). The number of mast cells and the percentage of mast cells with degranulation in middle - dose and high - dose CP groups were obviously decreased as compared with model group ( P 〈 0.05). Conclusion The middle - and high - dose CP can decrease the sensitivity of intestinal tract and relieve the degranulation of mast cells in IBS rats model.
