摘 要:目的探讨PPARγ和MMP-2与早孕细胞滋养细胞浸润的关系;方法本研究通过免疫组织化学技术检测早孕早期和早孕晚期绒毛组织中PPARγ和MMP-2蛋白的表达;结果PPARγ蛋白主要定位在早孕绒毛细胞滋养细胞核及绒毛外滋养细胞柱和细胞岛的细胞滋养细胞核内,MMP-2阳性颗粒主要存在绒毛细胞滋养细胞和合体细胞滋养细胞胞浆内,绒毛间质细胞内亦有阳性染色,且早孕早期绒毛PPARγ表达量高于早孕晚期(P<0.05),而MMP-2表达量在早孕晚期高于早期(P<0.05),与PPARγ呈负相关(r=-0.74,P<0.01);结论PPARγ在调节滋养细胞浸润过程中起重要作用,而且其作用可能是通过调节MMP-2的表达实现的,为深入探讨PPARγ及配体在滋养细胞浸润机制中的作用提供实验基础。Objective The objectives of our current study were to investigate the relation of PPARγ and MMP-2 with trophoblast invasion. Methods Our research was detected the expression of the protein of PPARγ and MMP-2 in early and late first trimester pregnancy by immunohistochemistry. Results PPARγ protein is located manly in in the nuclei of cytotrophoblasts from proximal and distal columns of the anchoring villi and villous cytotrophoblast, MMP-2 is located manly in the intracytoplasm of cytotrophoblast and syncytiocytotrophoblast and the villous stromal core as well. The expression of PPARγ were higher in early first trimester than in late first trimester(P〈 0.05 ), and the expression of MMP-2 more in late first trimester than in early first trimester (P〈0.05), there is the negative correlation between PPARγ and MMP-2 (r=--0.74, P〈0.01 ).Conclusion PPARγ play an important role in the modulation of thophoblast invasion, PPARγ ligands can inhibit invasion through downregulate MMP-2 dxpressions, these provided experiment basement for deeply investigation action of PPARγ ligands in the mechanism of cytotrophoblast invasion.
