摘  要:[目的]了解新生期注射己烯雌酚(diethylstilbestrol,DES)后BALB/c小鼠脾细胞黄体生成素(luteinizinghormone,LH)表达的变化以及这种变化在不同性别之间是否存在差异。[方法]选取同一窝、体重差异小于5%的新生BALB/c小鼠,随机分为对照组和DES组,雌、雄各选25对,在生后24h内,DES组小鼠颈背部皮下注射DES,每隔24h注射1次,共连续注射5次。对照组小鼠平行注射等量无菌豆油。分别在生后d7、14、21、35和d49将小鼠处死,取其脾组织进行常规石蜡包埋、切片及LH的免疫组织化学检测。[结果]无论是雌性小鼠还是雄性小鼠,从生后d7到d49,对照组和DES组小鼠脾内均可见LH阳性细胞,阳性细胞呈淋巴细胞和小鼠脾巨噬细胞形态结构特点,阳性表达在两组均随周龄的增加而减弱;同一时间点条件下观察发现,DES组的LH阳性表达均低于对照组。雌性小鼠和雄性小鼠相比,雌性小鼠在各时间点DES组脾内的LH阳性细胞数量均低于对照组,且两组之间差异具有统计学意义;而雄性小鼠的DES组在生后d7和d14与对照组之间无差异,在生后d21、35和d49与对照组相比差异具有统计学意义。[结论](1)无论是雌性还是雄性BALB/c小鼠,其脾内均存在LH表达细胞,表达水平随年龄的增加而降低;(2)新生期接触DES可导致BALB/c小鼠脾内LH阳性细胞减少,减少程度与小鼠的性别有关,雌性小鼠对DES的敏感性大于雄性小鼠。(3)不论是雌性小鼠还是雄性小鼠,DES对其脾内LH表达的抑制作用均可从新生期持续到成年期。[Objective] To investigate the dynamic changes of expression of luteinizing hormone (LH) in spleen of BALB/c mice after treated with diethylstilbestrol (DES) in neonatal period and whether these changes differ from female mice to male mice. [Methods] Neonatal brood BALB/e mice whose weight discrepancy was less than 5% were divided randomly into control group and DES group, with 25 pair of female mice and 25 pair of male mice respectively. Within 24 hours after birth mice in DES group were injected subcutaneously on cervix-backside with DES at intervals of 24 hours for 5 times, and mice in control group were injected with germfree bean oil with the same method. Mice were killed on the day 7, 14, 21, 35 and 49 after birth separately, and the spleen was taken out, imbedded with paraffin and sectioned serially in 5 μm. The expression of LH in slice of spleen was detected by immunohistochemistry technique. [ Results] No matter in female mice or male mice, LH positive cells were all present in both DES-treated group and control group from day 7 to day 49. Positive cells were recognized as lymphoey'tes and macrophages according to their morphological characteristics. Age-dependent decrease of positive expression of LH was showed in both groups. In the same time point, the expression of LH in DES group was lower than that in control group. However, the extent of decrease was different between the two sexes. In female mice, a significant statistic difference in amount of LH positive cells in spleen between DES group and control group was observed from day 7 to day 49, While in male mice, the significant statistic difference was only be observed on day 21, 35and 49. [Conclusion] ( 1 ) LH expression exists in spleen of both sexes of BALB/c mice and decreases gradually with the increase of age of mice. (2) The contact of DES in neonatal period can lead to a decreased LH expression in spleen of BALB/c mice, and the extent of decrease is relative to the sex of mice.
