血管紧张素转换酶抑制剂与醛固酮受体拮抗剂对腹膜纤维化的比照研究

摘　　要：目的:探讨腹膜纤维化的防治措施。方法:40只雄性Wistar大鼠随机分成4组,A组腹腔内每日注入20毫升4.25%百特透析液;B组腹腔内每日注入20毫升4.25%百特透析液,同时血管紧张素转换酶抑制剂一平苏10 mg/kg.day管喂;C组(10只)腹腔内每日注入20毫升4.25%百特透析液,同时醛固酮受体拮抗剂安体舒通100mg/kg.day管喂。D组(10只)正常对照组,每日腹腔内注入生理盐水20毫升。ABC三组从试验开始时于试验第1、3、5、7天按大鼠体重给于腹腔注射脂多糖(LPS)0.6 mg/kg,并于第1天测定腹透液中的转化生长因子-β。30天后收集大鼠腹膜作病理,并测定腹透液中的转化生长因子-β。结果:给药组(B,C组)的腹膜病理改变比模型组(A组)轻,转化生长因子-β浓度也明显下降,具有统计学意义。结论:血管紧张素转换酶抑制剂及醛固酮受体拮抗剂能有效的防治腹膜纤维化的进程。Objective：To investigate the prevention and treatment of peritoneal fibrosis in long-term peritoneal dialysis patients.Methods：Forty male Wistar rats were divided into four groups,Group A（n=10）,B（n=10）medicine group,angiotensin-converting enzyme inhibitor（inhibace） 10 mg/kg/day was given orally to rats by gastric gavage in the morning once a day.C（n=10） medicine group Aldosterone receptor blocker.100 mg/kg·day was given orally to rats by gastric gavage in the morning once a day.D（n=10）control group,received no treatment.Groups A B and C received daily intraperitoneal injection of 20 mL 4.25% Beter dialysate,on day 1,3,5,7,these rats was given intraperitoneal injection of LPS 0.6 mg/kg.Thirty days later all rats were sacrificed.TGF-β expression were examined in abdominal cavity liquid of peritoneal dialysis rats.Histopathology of parietal peritoneum was examined.Results：TGF-β obviously increased in Both A B and C groups as compared to D group（P〈0.01）,group A is more serious than group B and C（P〈0.01）,so do histopathology of peritoneum.Conclusion：Both Aldosterone receptor blocker and angiotensin-converting enzyme inhibitor are benefit to the prevention and treatment of peritoneal fibrosis.
