重组人肿瘤坏死因子-α对人正常精子运动影响的实验研究

摘　　要：目的观察重组人肿瘤坏死因子-α（rhTNF—α）在体外对人正常精子运动的影响。方法40例健康自愿者正常精液，Peroll梯度离心将精子密度调整为10×10^6／ml，分别与对照组以及终浓度为30pg／ml、60pg／ml、90pg／ml、270pg／ml的rhTNF-α孵育。在0．5h、1h、2h、3h、4h时间段取样，用CASA检测精子运动参数变化。结果rhTNF—α终浓度为60、90、270pg／ml各组精子的活力、直线速度、曲线速度、平均速度和前向运动明显低于对照组，差异均有统计学意义（P〈0．01），且270pg／ml组的rhTNF—α对精子作用最为显著。结论一定浓度的rhTNF—α可明显抑制人正常精子运动。Objective To study the spermatozoon motility in vitro. Methods effects of recombinant human tumor necrosis factor （rhTNF- α ）on the Forty semen samples from healthy men were collected by masturbation. sperm were seperated by Percoll gradient centrifugation and was suspended to a concentration of 10 × 10^6/ml in Ham＇s F10 medium, and they were incubated with TNF- α at the final concentration of 0pg/ml（control group）, 30pg/ ml, 60pg/ml, 90pg/ml, 270pg/ml respectively. Semen were collexted and analyzed by CASA at time point of 0.5h, lh, 2h, 3h, 4h. Results The motility, viability,straight linear velocity, curvilinear velocity, average path velocity, progressive motility of sperm in test groups（30pg/ml, 90pg/ml, 270pg/ml TNF- α ） was lower than those in control group（0pg/ml TNF- α ）（P〈0.01）. especially at the concentration of 270pg/ml, effect on human sperm was remarkable. Conclusion Sperm motility was obviously inhibited by rhTNF-α with defined concentration
