摘　　要：目的观察人工合成的两种CpG寡核苷酸(CpG ODN)对乙型肝炎疫苗免疫小鼠淋巴细胞增殖效应的影响。方法BALB/c小鼠分别经乙肝疫苗、乙肝疫苗+CpG1826和乙肝疫苗+CpG2006免疫后,取小鼠脾和胸腺淋巴细胞,进行非特异性和特异性淋巴细胞增殖试验,应用MTT法分别检测淋巴细胞增殖效应。结果CpG两组与疫苗组及对照组相比,脾淋巴细胞增殖效应均显著增强,CpG1826组均优于CpG2006组,且差异有统计学意义;CpG两组的胸腺淋巴细胞非特异性增殖效应也显著增强,但CpG两组之间差异无统计学意义。结论CpG1826和CpG2006均能明显刺激BALB/c小鼠脾脏及胸腺淋巴细胞增殖,且CpG1826显示出更强的脾淋巴细胞免疫刺激效应。Objective To observe two kinds of synthetic CpG oligodeoxynucleotide （ODN） on the lymphocyte proliferation in mice immunized with hepatitis B （HB） vaccine. Methods Divide BALB/c mice into 4 groups randomly. The mice in 3 test groups were immunized with HB vaccine, HB vaccine ＋ CpG 1826 and HB vaccine ＋ CpG 2006 respectively, and those in control group with physiological saline. Take the spleen and thymus of immunized mice for specific and non-specific lymphocyte proliferation tests. De- termine the proliferation level of lymphocytes by MTT method. Results The proliferation level of spleen lymphocytes was significantly higher in HB vaccine ＋ CpG 1826 and HB vaccine ＋ CpG 2006 groups than in HB vaccine and control groups, and significantly higher in HB vaccine ＋ CpG 1826 groups than in HB vaccine ＋ CpG 2006 group. Though the non-specific proliferation levels of thymus lymphocytes in HB vaccine ＋ CpG 1826 and HB vaccine ＋ CpG 2006 groups showed no significant difference, both of them were significantly higher than those in HB vaccine and control groups. Conclusion Both CpG 1826 and CpG 2006 stimulated the proliferations of lymphocytes in spleen and thymus of mice significantly. However, CpG 1826 showed strong immunostimulating effect on the proliferation of spleen lymphocytes.
