摘 要:目的探讨腹腔注射硫酸锌(ZnSO4)诱导热休克蛋白(HSP70)合成及其对大鼠移植胰腺再灌注损伤的保护作用。方法应用同系大鼠异位全胰十二指肠移植模型,移植术前和术后5 min大鼠腹腔注射ZnSO4[锌离子(Zn2+):5 mg/kg(Zn-1组),10 mg/kg(Zn-2组),15 mg/kg(Zn-3组)]。移植术后24 h经腹主动脉取血测定大鼠空腹血糖、血清脂肪酶和淀粉酶含量,测定移植胰腺组织中HSP70含量(免疫印迹法)及髓过氧化酶(MPO)活性,并观察组织学变化。结果移植术后24 h Zn-1组,Zn-2组,Zn-3组移植胰腺HSP70含量均较术前明显升高(均为P<0.05),3组间差异无显著性(P>0.05);移植后24 h血糖、血清脂肪酶和胰腺组织中MPO水平:Zn-2组<Zn-1组<移植对照组<Zn-3组,差异均有显著意义(P<0.05)。Zn-2组和Zn-1组胰小叶间质水肿和胰小叶内中性粒细胞浸润较轻。结论外源性应用恰当剂量的ZnSO4(Zn2+5 mg/kg和10 mg/kg)能增加胰腺组织HSP70含量,对移植胰腺再灌注损伤起保护作用;剂量过大则其保护作用消失。Objective To study the protective effects of the heat shock protein 70 ( HSP70 ) induced by zinc sulfate on reperfusion injury following pancreaticoduodenal transplantation in rats. Methods The homologous male Wistar rat model o( heterotopic total pancreaticoduodenal transplantation was used. The ZnSO4 treated rats received the intravenous injection of Zn2+ 5 rains before and after operation at a dose of 5 mg/kg ( Zn-1 group) ,10 mg/kg (Zn-2 group) and 15 mg/kg (Zn-3 group), and the control group with the same volume of saline. The tissue concentration of HSP70 was determined using Western-Blot. In addition, blood sugar (BG) and serum concentration of amylase and lipase were examined 24h after transplantation, and the activity of myeloperoxidase (MPO) in the pancrease graft was measured at the same time. Histological observation was performed. Results Light microscopic studies showed that histomorphological changes of pancreas in Zn-2 group and Zn-1 group were much less than those in control group and Zn-3 group. The value of BG and serum lipase and MPO in Zn-2 group 〈 Zn-1 group 〈 control group 〈 Zn-3 group(P 〈 0.05). Conclusions Zn2+ (at the dose of 5 mg/kg and 10 mg/kg) can increase the amount of HSP. Zn^2+ has protective effects on the ischemia/reperfusion injury of pancreatieoduodenal transplantation, but the effect will disappear when the dose is over high.
