摘 要:目的:研究新结构类型钾通道开放剂埃他卡林新衍生物对大鼠离体工作心脏功能的影响.方法:在大鼠离体工作心脏上,以吡那地尔为阳性对照药,分别给予15个埃他卡林新衍生物(浓度为1,10,100μmol/L)灌流,用八导生理记录仪记录心脏功能参数,连续30min观察其对心脏功能的影响.结果:所有15个化合物在3种浓度水平下,以化合物3为例,30min内对心脏功能参数HR,LVSP,+dp/dtmax,-dp/dtmax均未见明显影响(P>0.05).结论:埃他卡林新衍生物对离体工作心脏功能无影响,提示其不通过直接影响心脏功能而产生降压作用.AIM: To study the effects of new derivatives of iptakalim, a novel potassium channel opener on the functions of isolated working hearts of rats. METHODS: During a period of 30 min after administration of 15 compounds at the concentrations of 1, 10, 100 ~mol/L in isolated working hearts of rats with pinacidil as comparative drug, we observed and recorded parameters of cardiac function by polygraph with eight channels in order to evaluate the pharmacological effects of those compounds. RESULTS:Compound 3 as well as all the rest of 15 compounds did not significantly change the parameters of cardiac functions of isolated working hearts of rats(P 〉 0.05 ). CONCLUSION : The new derivatives of iptakalim have no significant effects on cardiac functions of isolated working hearts of rats, which suggests that they don't lower blood pressure through the direct effect on heart.
