摘  要:目的:小热休克蛋白αB-晶状体蛋白(αB-crystallin)在骨骼肌有丰富的表达,但其在运动训练中的准确功能有待证实,通过观察αB-crystallin和骨架蛋白desmin在离心运动训练中含量的变化,阐明αB-crystallin与骨架蛋白的关系。研究方法:实验使用2月龄雄性Wistar大鼠56只,电动跑台,间歇性离心运动训练方案,分为安静对照组和离心训练3周组,离心训练6周组,之后安静对照组和训练6周组做大强度离心运动;取大鼠腓肠肌,采用蛋白免疫印迹法分析αB-晶体蛋白含量变化,采用免疫组织化学法分析骨架蛋白desmin变化。结果:6周的间歇性离心训练使腓肠肌αB-晶状体蛋白有显著的增加,训练鼠在进行大强度离心运动时,表现出对训练的适应,骨架蛋白desmin丢失减少,CK显著的降低,说明增加的αB-晶状体蛋白对骨架蛋白有保护作用;而未训练大鼠在大强度离心运动中体验了一个显著的应激反应,骨架蛋白desmin显著丢失,αB-crystallin有显著增加。结论:小热休克蛋白αB-晶状体蛋白在离心运动引起的骨架蛋白损伤中可能是一种补充保护作用机制。Objective: Small heat shock protein(αB-crystallin) is constitutively expressed in skeletal muscle,But their precise functions in the exercise remain largely undefined.To clarify the relationship between αB-crystallin and desmin,we have studied the concentration of αB-crystallin and desmin in muscle after eccentric exercise.Method: Fifty-six 2-mo-old male wistar rats to run on a motorized treadmill are randomly assigned to sedentary control group(Sed),3 weeks eccentric training group(3ET),6 weeks eccentric training group(6ET),then 6ET and Sed performes a high intensity eccentric exercise.Western bloting is used to analyze the expression of αB-crystallin in gastrocnemius.Immhistochemistry is used to analyze the information about loss of desmin in gastrocnemius.Result: 6 weeks intermittent eccentric training induces gastrocne-mius αB-crystallin expression significantly.Both untrained and trained rats are submitted to the same bout of high intensity eccentric exercise.Trained rats clearly demonstrated higher resistance to exercise,such as desmin disruption is less and CK is decrease significant,which suggest αB-crystallin protect cytoskeletal from exercise stress.In high intensity eccentric exercise,untrained rats experience a significant stress response,a significant increase in αB-crystallin levels parallel to a decrease in the desmin content.Conclusion: αB-crystallin acts as complementary protective mechanism in eccentric exercise-induce cytoskeletal injury.
