摘  要:目的:研究新型雌激素17aα-D-高炔雌二醇-3-乙酯(DHEA)的抑瘤活性。方法:对DHEA进行肺腺癌细胞系A549、肝癌细胞系Bel-7402的体外抑瘤试验,计算半数抑制浓度(IC50);在小鼠体内进行宫颈癌细胞U14抑瘤实验及与γ-射线协同作用的肉瘤细胞S180的抑瘤增效实验,计算抑瘤率。结果:体外试验中DHEA对A549和Bel-7402的IC50分别为12.28、17.79μg·mL-1;体内实验中对U14抑瘤率最高达64.3%,对S180抑瘤率合用及DHEA单用的抑瘤率均高于50%,且合用组更高。结论:DHEA具有体内、外抑瘤活性,且与γ-射线合用具有协同增效作用。OBJECTIVE: To study the anti - tumor activity of 17aα - D - homo - ethynylestradiol - 3 - acetate (DHEA), a neotype estrogen. METHODS: An in vitro tumor - inhibition test of DHEA on carcinoma cell line A548 and Bel - 7402 was conducted and the IC50 were computed ; meanwhile an in vivo tumor - inhibition test of DHEA on U14 and a test of its synergic action with γ - rays on S180 inhibition in mice were conducted, and the tumor - inhibition rate was computed. RESULTS: The IC50 of DHEA in vitro against A549 and Bel - 7402 cells were 12.28 and 17.79ug · mL^-1, respectively. In vivo test showed that the inhibition rate on U14 was as high as 64.3%, and the inhibition rates on S180 were all higher than 50% in mice treated with DHEA alone or in combination with γ- rays radiation, higher in the combination therapy group than in single DHEA group. CONCLUSIONS: DHEA showed anti- tumor activities both in vivo and in vitro as well as synergistic action with γ- rays on tumor - inhibition.
