摘 要:目的:研究两种不同强度耐力训练后力竭运动大鼠肾细胞凋亡的差别及其机制。方法:大鼠随机分为对照组(C组)、低强度和大强度耐力训练后力竭组(LE组、HE组),经5周以上耐力训练后进行一次力竭运动,24h后取材。透射电镜和原位末端标记法检测肾细胞凋亡,免疫组化法检测肾细胞Bcl-2和Bax蛋白表达,硝酸还原酶法检测肾组织NO生成,测定NOS活性。结果:与C组比较,HE组肾小管上皮细胞凋亡显著增加,LE组凋亡不明显;各组Bax蛋白表达差异不显著;LE组Bcl-2蛋白表达明显增高,NOS活性显著降低,而HE组无显著变化,NO浓度基础值最高。结论:低强度耐力训练能增高Bcl-2蛋白表达、降低NOS活性以稳定肾组织NO含量,控制力竭运动诱导的细胞凋亡;耐力训练可能对大强度力竭运动明显诱导肾细胞凋亡发挥作用,但不能起到完全控制效果。Objective:to study the difference and possible mechanism of renal cells apoptosis in two kinds of intensity exhausting exercise after enduranced-training increasing by degrees. Methods: rats were randomly divided into control group (C group), low intensity endurance and exhausting exercise group (LE group), high intensity endurance and exhausting exercise group (HE group). The LE group and HE group ran with an exhausted exercise after five weeks of different intensity endurance-training. The authors sampled kidney tissue at 24 hours after exhausting exercise. The apoptosis, Bcl-2 and Bax protein expression of renal cell was investigated with TUN-EL and immunohistochemistry method. The content of NO was detected by nitric acid reductase. NOS activity was reflected by detecting nmol of NO in tissue protein per unite weight during unit time. Results: in the LE group, the kidney apoptosis didn't increase while it increased very significantly in the HE group. Bax protein expression were insignificantly different among three groups. In the LE group Bcl-2 protein expression of renal cells increased very significantly, the NOS activity decreased. The NO content of the HE group trended to increase, the NOS activity didn't change significantly. Conclusion: the results suggest that low intensity endurance-training can restrain renal cells apoptosis induced by exhausting exercise with increasing Bcl-2 protein expression and leveling off NO content. Endurance-training may protect the kidney cells and decrease the kidney cells apoptosis , but it can't control the apoptosis completely.
