重组白细胞介素-2对血脑屏障结构及功能的影响

摘  要:采用辣根过氧化物酶(HRP)作为示踪剂,观察静脉注射重组白细胞介素-2(rIL-2)后血脑屏障通透性的变化。结果发现:注射rIL-2后血脑屏障对HRP的通透性明显增加,而脑毛细血管内皮细胞结构完整。电镜结果提示这种变化与毛细血管内皮细胞的转运功能增强有关。Blood-brain barrier status and cerebral vascular structure integrity in rats after human recombinant interleukin-2 (rIL-2) infusion were examined. Horseradish peroxidase CHRP) was used as permeability probes to assess cerebrovascular permeability. Eighteen Wistar rats were divided into two groups. In the first group, each of nine animals received both HRF (50mg/kg) and rIL-2 (100 000 units/kg). Animals in the second group were injected HRP and sterile saline (the same volume as rIL-2). Four animals in each group were sacrificed 2h postinfusion in order to assess initial change,the others were sacrificed at 12h after injection. Brain sections were examined with light microscopy and electronmicroscopy. The results suggested that cerebrovascular permeability be increased following rIL-2 infusion,but blood-brain barrier integrity has no change. It demonstrated that rIL-2 may increase the active transport of cerebrovascular endothcliocyte.
