摘 要:目的:考察活血化瘀中药成分抗肿瘤-血管内皮细胞黏附能力及其机制。方法:运用体外荧光标记肿瘤-内皮细胞黏附模型对中药成分抗肿瘤细胞黏附能力进行考察,并运用Western blotting实验手段检测相关黏附分子蛋白表达。结果:对几种活血化瘀中药成分进行筛选后发现大黄素具有一定的抗肿瘤-内皮黏附能力,同时可降低内皮细胞黏附分子ICAM-1及E-selectin的表达。结论:大黄素具有抗肿瘤-内皮细胞黏附能力,可能是通过降低黏附分子表达的机制实现的。Objective: To investigate the anti-adhesion effect of Chinese medicine extracts effective in activating blood and removing stasis on tumor endothelial cells and its mechanism. Methods: The model was established with the MDA-MB-231 breast cancer cells and monolayers of human umbilical vein endothelial cells (HUVEC) in vitro. The tumor cells were specifically labeled with a fluorescent dye for quantification. For increasing the amount of adherent cells, HUVEC monolayers were stimulated with phorbol ester(TPA) before the addition of the tumor ceils. To investigate the possi ble mechanism of Emodin's anti-adhesion tumor cells to endothelial cells, the expression of adhesion molecules ICAM-1, VCAM-1, E-selectin were studyed with Western blot assay. Results: Among several extracts tested, Emodin reduced tumor cell's adhesion to HUVECs to 89.50 % at 1μg/mL, 78.13 % at 2 μg/mL and 77.12 % at 6μg/mL, and the expression of CAM-1 and E-selectin. Conclusion: Emodin has the effect of reduceing tumor cell's adhesion to HUVECs by down-regulating the expression of adhesion molecules.
