摘 要:[目的]研究微波萃取法提取盾叶薯蓣皂苷元的最佳条件。[方法]采用微波萃取法提取盾叶薯蓣皂苷元,高效液相色谱法检测薯蓣皂苷元提取率,选取粉碎粒度、提取时间、提取温度、固液比为影响因素进行单因素试验和正交试验,确定盾叶薯蓣中皂苷元的最佳提取工艺。[结果]单因素试验发现石油醚作提取溶剂、粉碎粒度为60目、固液比为1∶20(g/ml)、提取时间为10 min、提取温度为100℃,盾叶薯蓣中皂苷元的提取率最高。正交试验表明,微波萃取法提取盾叶薯蓣皂苷元的最佳条件为:粉碎粒度60目,提取时间10 min。[结论]在最佳提取条件下微波提取盾叶薯蓣皂苷元,提取率达1.837%。[Objective] The research was to study the optimum extraction conditions for diosgenin from Dioscorea zingiberensis by microwave extraction method.[Method] The microwave extraction method was taken to extract the diosgenin from D.zingiberensis,and the extraction rate of diosgenin from D.zingiberensis was detected by high performance liquid chromatography(HPLC).The factors of grinding particle size,extraction time,extraction temperature,solid-to-liquid ratio were taken as the effecting factors to make single factor test and orthogonal test and the optimum extraction technology for diosgenin from D.zingiberensis was determined.[Result] The single factor test showed that when petroleum ether was used as extraction solvent,grinding particle size was 60 meshes,solid-to-liquid ratio was 1∶20(g/ml),extraction time was 10 min and extraction temperature was 100 ℃,the extraction rate of diosgenin from D.zingiberensis was the highest.The orthogonal test showed that the optimum condition for extracting diosgenin from D.zingiberensis by microwave extraction method was grinding particle size of 60 meshes,extraction time of 10 min.[Conclusion] Under the optimum extraction condition,the extraction rate of diosgenin from D.zingiberensis by microwave extraction method reached 1.837%.
