摘 要:[目的]从湖水中分离小球藻并提取其DNA。[方法]采用稀释法及水滴法分离小球藻,优化小球藻培养条件,并用改良CTAB法及SDS法提取其DNA。[结果]温度20~25℃、光照4.39~5.86W/m^2及转速100~150r/min的条件适于小球藻的培养;改良CTAB法适于提取小球藻DNA。[结论]该研究将有助于在分子水平研究小球藻,促进小球藻的开发和利用。[Objective]The aim of this study is to isolate Chlorella vulgaris (chlorella) and extract its genomic DNA. [Method] Both the dilution method and drip method were employed to isolate chlorella from lake water samples; the conditions for culturing chlorella were optimized and its genomic DNA was extracted by improved CTAB method and SDS method. [Result] The proper conditions for chlorella culture were as following: temperature 20 -25℃, illumination 4.39-5.86 W/m^2 and rotational speed 100-150 r/min; improved CTAB method was suitable for extracting genomic DNA from chlorella. [ Conclusion] The result is helpful for studying the chlorella at molecular level and promote the exploitation and utilization of chlorella resources.
