脂多糖致感染性脑水肿大鼠脑组织TLR4 mRNA表达的变化

摘　　要：目的:观察脂多糖(LPS)致感染性脑水肿模型大鼠脑组织中TLR4 mRNA表达的动态变化,探讨TLR4在感染性脑水肿中的作用。方法:1月龄SD大鼠25只,随机分为2组。对照组(5只)颈内动脉注射0.1 mL生理盐水,颈外静脉注射伊文思蓝(EB),6 h后断头处死;LPS组(20只)颈内动脉注射100μg LPS,颈外静脉注射EB,分别于注射6、12、24、48 h后各取5只大鼠断头处死。取脑,用干湿重法测定脑组织含水量,甲酰胺法测定EB含量,RT-PCR法检测脑组织TLR4 mRNA的表达量。结果:LPS注射后大鼠大脑血管周围间隙增宽,炎性细胞浸润;胶质细胞肿胀,神经元周隙增宽。对照组无上述水肿表现。LPS注射6 h后大鼠脑组织含水量、EB含量升高,TLR4 mRNA表达降低;12 h时脑组织含水量、EB含量升高达峰值,24 h、48 h时逐渐降低至正常水平;注射LPS 12 h后,脑组织TLR4 mRNA表达量降至最低,24 h和48 h时逐渐恢复。大鼠脑组织EB含量、含水量与TLR4 mRNA表达量均呈负相关(r分别为-0.604,-0.535,P均<0.05)。结论:TLR4在脑水肿发展过程中可能有特殊的保护作用。Aim： To observe the change of TLR4 mRNA exprssion in the brain tissue of infant rats with infectious brain edema induced by lipopolysaccharide（LPS）.Methods：A total of 25 healthy and 1 month-old SD rats were randomly divided into two groups.Normal saline group（NS group,n=5）,0.1 mL normal saline was injected by carotid for each rat,and the rats were decapitated after 6 h.LPS group（n=20）,each rat was applied 100 μg LPS,and 5 rats were decapitated after 6 h,12 h,24 h,48 h,respectively.The water content of brain tissue was determined by measuring both wet weight and dry weight.The EB content of brain tissue was determined by the methods of formamide and RT-PCR was used to detect the expression of TLR4 mRNA.Results： The pathological characteristic of brain edema appeared in the brain tissues of the rats of LPS group.The water content and EB content in brain tissues of LPS 6 h group increased at the 6th hour after LPS injection,and reached to the peak at the 12th hour,and then decreased.The change of TLR4 mRNA expression was contrary.The expression of TLR4 mRNA was negatively correlated to the water content and EB content in brain tissue（r was-0.535,-0.604,respectively,P〈0.05）.Conclusion：TLR4 may play a protective role during the development of infectious brain edema.
