摘 要:目的探讨一氧化氮对慢性缺氧大鼠肺动脉内皮细胞及平滑肌细胞内皮素-1(ET-1)mRNA表达的影响,全面了解缺氧性肺动脉高压的发病机制。方法对经L-精氨酸(L-Arg)及Nω-硝基-L-精氨酸甲酯(L-NAME)处理的慢性缺氧大鼠(40只)肺组织,以ET-1cRNA探针进行原位杂交。结果缺氧1周及缺氧2周大鼠大部分肺动脉(75%±3%及71%±6%)其1%~50%的内皮细胞显示ET-1mRNA杂交阳性信号。L-Arg使大鼠肺动脉内皮细胞ET-1mRNA表达明显抑制,而L-NAME使其表达明显增强。缺氧1周及缺氧2周大鼠大部分肺动脉(85%±6%及98%±2%)的平滑肌细胞无ET-1mRNA表达,但L-NAME使其表达增强。结论一氧化氮对慢性缺氧大鼠肺动脉内皮细胞及平滑肌细胞ET-1-mRNA表达均有抑制作用。Objective\ To investigate the impact of nitric oxide on endothelin 1(ET 1)mRNA expression in pulmonary artery endothelial cells and pulmonary artery smooth muscle cells of chronic hypoxic rats.Methods\ In situ hybridization was performed on lung sections from40chronic hypoxic rats treated either with L Arginine(L Arg)or N ω nitro L arginine methyl ester(L NAME)by using cRNA probe for ET 1. Rresults\ Most intrapulmonary arteries had1%~50%of the endothelial cells expressing ET1mRNA in both one week and two week hypoxic rats(75%±3%and71%±6%,respectively), which was significantly inhibited by L Arg but augmented by L NAME administration.Most pulmonary artery smooth muscle cells showed no ET 1mRNA signals in both one week and two week hypoxic rats(85%±6%and98%±2%,respectively).However,L NAME increased ET 1mRNA expression in pulmonary artery smooth muscle cells of hypoxic rats.Conclusion Nitric oxide Inhibited ET gene expression in both pulmonary artery endothelial cells and smooth muscle cells of rats exposed to chronic hypoxia.
