摘 要:【目的】探讨骨髓间质干细胞(MSC)对同种大鼠骨髓移植后移植物抗宿主反应(GVHD)的影响,尤其是MSC对脾脏结构重建的作用。【方法】建立Fischer344(RT-1Al)大鼠至Wistar(RT-1Au)大鼠的同种异体骨髓移植模型,观察移植后大鼠的基本生活情况,病理学分析靶器官皮肤、肝脏、肠道和脾脏的组织学改变,尤其注意MSC对同种骨髓移植后脾脏结构重建的影响。【结果】①在移植后30d的观察期内,共移植组显示较高的生存率。②组织学分析显示:在共移植组,靶器官皮肤、肝脏、肠道的损伤相对较轻。③脾脏组织学分析显示:在骨髓与MSC共移植组,受体脾脏结构接近正常;在单纯骨髓移植组,受体脾脏白髓萎缩,动脉周围淋巴鞘只有少量T细胞,无典型脾小结形成。【结论】MSC与骨髓共移植对GVHD有抑制作用,并显著促进脾脏结构重建。[Objective] To investigate the effects of mesenchymal stem cells (MSC) on severity of graft vs host disease (GVHD) after allogeneic bone marrow transplantation (allo-BMT), especially on structural remodeling of spleen which was known as important peripheral immune organ and the GVHD target organ. [Method] BMT model from Fischer3d4 rats (RT-1A1) to Wistar rats (RT-1Au) was established. The rats' basic living conditions and the pathological changes of target organs such as the skin, the liver and the intestinal tract, as well as the reconstitution of splenic architecture especially were analyzed. [Results ] It was demonstrated that the cotransplantation group had a higher survival rate at 30 days postransplant. The histology analysis showed that the skin, the liver, and intestinal tract in cotransplantation group were damaged relatively slightly. By histologic analysis, the spleens from cotransplantation group appeared nearly structural normal, with well-developed red and white pulp; while the spleens from BM transplantation group were atrophic, containing less T cells forming periarteriolar lymphoid sheaths (PALS),without distinct follicles. [ Conclusions ] Cotransplantation of MSC and the bone marrow could suppress GVHD, and remarkably promote reconstitution of splenic architecture.
