摘 要:在比较了植物DNA数种提取方法之后,提出了一种改良的DNA提取方法,可以快速地从沙漠植物中提取到高质量的DNA样品,降低了实验成本。紫外吸收的测定结果表明:所获DNA的A260nm/A280nm值大于1.75。琼脂糖凝胶电泳结果表明:DNA片段的大小为50kb左右。用RNA酶去除残留RNA的效果良好,限制性酶切实验合格。该方法对于其他植物材料也非常有效。In this paper, an improved arid and desert plants DNA isolation method was suggested based on the comparison among several different methods. Higher qualitative DNA samples could be obtained with this method, and the experimental steps and costs could be decreased. The results of U.V absorption showed that the value of the A260 nm/A 280 nm of DNA samples>1.75;The results of agrose gel electrophoresis showed that the fragments of DNA samples are about 50 kb;The remains of RNA in the DNA samples coud be removed by RNase treating. The restriction test was qualified. This method is also quite effective for other materials.
