p38丝裂原活化蛋白激酶抑制剂对大鼠骨性关节炎的影响

摘　　要：目的观察p38丝裂原活化蛋白激酶抑制剂SB203580对大鼠膝关节骨性关节炎关节软骨的影响。方法40只SD大鼠随机分为A、B、C、D 4组。A、B、C组行单侧膝关节前交叉韧带切除术,A组于术后行关节腔内注射0.1 mL浓度为100μm/L SB203580,B组注射等量生理盐水做为实验对照,C组不予任何处理为空白对照组,D组为正常对照组。术后8周处死动物。观察各组标本大体评分、Mankin评分、软骨细胞凋亡指数以及软骨Ⅱ型胶原免疫组化染色。结果40只大鼠均纳入结果分析。大体评分及Mankin评分显示A组软骨退变明显轻于B、C组(P<0.05);各组均发现有凋亡的软骨细胞,A组软骨细胞凋亡指数低于B、C组(P<0.05),D组软骨细胞凋亡指数明显低于其他3组(P<0.05);A组关节软骨Ⅱ型胶原染色吸光度值大于B、C组(P<0.05)。结论p38MAPK抑制剂对关节软骨有一定的保护作用,可以延缓OA进程,对OA有一定的治疗作用。Objective To observe the influence of p38 mitogen activated protein kinase inhibitor SB203580 on the cartilage in rat osteoarthritis. Methods 40 SD rats were randomly divided into group A, B, C, and D. Group A, B, and C underwent unilateral anterior eruciate ligament transection. Rats of group A received 0.1 mL intraarticular injection of SB203580 at concentration of 100 μm/L immediately after surgery, while those in group B received 0.1 mL 0.9% Sodium Chloride. Rats of group C received no treatments and those of group D were normal. At the 8th week after surgery, the rats were sacrificed. Macroscopic and histologic studies, chondrocyte apoptosis index, type H collagen of cartilagewere observed and recorded. Results All of the 40 SD rats were analyzed. Histological changes observed in group A were slighter than those in group B and C （ P 〈 0.05 ） ; Apoptotic chondrocytes were observed in all of those groups, and apoptosis index of group A was lower than group B and C（ P 〈 0.05）, and rats in group D had much lower apoptosis index than the other three groups（ P 〈 0.05） ; More content of type I] collagen in group A were observed than in group B and C （ P 〈 0.05）. Conclusion The p38 MAPK inhibitor can protect the articular cartilage and inhibit the development of OA.
