摘  要:为阐明来自不同寄主的五针松疱锈病菌的关系,了解其遗传多样性水平,并为今后研究该菌内的遗传变异和流行监控提供依据,该文采用RAMS(Random Amplified Microsatellite)标记对吉林、辽宁红松疱锈菌的4个居群和云南、四川华山松疱锈菌3个居群的遗传多样性进行了研究。从18条引物中筛选出5条对全部菌株进行了扩增,共检测到42个可重复的有效位点,其中多态位点有25个,多态位点比率为59.52%,Nei’s基因多样度指数H=0.2524,Shannon多样性信息指数Hsp=0.3627,基因流Nm=0.1365。分析结果表明:不同寄主来源疱锈菌群的遗传多样性高于同一寄主来源疱锈菌群;同一寄主来源的疱锈菌居群间存在遗传差异,但程度小于居群内。基于遗传距离,用NJ法对各居群间进行聚类分析的系统树显示,来自红松和华山松的疱锈菌形成独立的分支,相互间的遗传距离很大,说明两者在遗传上有显著的异质性。In order to clarify the relationship of five-needle pine blister rusts from different host plants, understand the genetic diversity of those rust populations and support the future research on genetic variation and epidemic monitoring of these fungi, the genetic diversity of seven populations of five-needle pine blister rust, from Pinus koraiensis in northeastern China's Jilin and Liaoning provinces and from P. armandii in southwestern China's Sichuan and Yunnan provinces, was assessed using RAMS markers. A total of 42 loci were detected using 5 primers, of these 25 loci were polymorphic. The average percentage of polymorphic loci was 59.52 %. The results showed that the Nei' s gene diversity was 0.252 4, Shannon' s information index was 0.362 7, and gene flow was 0.136 5, respectively. There was a higher genetic diversity of blister rust population from different host plants than from same hosts. There was genetic differentiation among population from the same host, but it was lower than that within population. NJ dendrogram of pine stem rusts populations based on Nei' s (1978) genetic distance shows that the pine blister rusts from P. koraiensis and P. armandii form isolated branch, and that genetic distance value between them is quite high, indicating that marked heterogeneity exists in the two groups of fungi.
