摘 要:目的:观察麝香乌龙丸对兔膝骨关节炎软骨组织病理形态学、基质金属蛋白酶1(MMP-1)及其抑制剂(TIMP-1)的影响,探讨麝香乌龙丸对关节软骨的保护作用是否与调节MMP-1及TIMP-1的表达有关。方法:30只日本大耳白兔随机分为3组:A正常对照组,B模型对照组,C麝香乌龙丸组。用1.6%木瓜蛋白酶膝关节腔内注射,建立兔膝骨关节炎模型,C组灌胃给药4周后,观察软骨组织形态学的变化及软骨中MMP-1、TIMP-1的表达。结果:C组软骨病理损害程度轻于B组(P<0.05),MMP-1表达明显低于B组(P<0.01),TIMP-1表达高于B组(P<0.01)。结论:麝香乌龙丸能减轻膝骨关节炎关节软骨退行性改变,其对关节软骨的保护作用可能与调节MP-1及TIMP-1的表达有关。Objective: To observe the effect of Musk-Aconite-Earthworm pill on pathomorphology and expression of MMP-1 and TIMP-1 of cartilage in rabbits with KOA, and to explore whether there is certain relation between cartilage protection and expression of MMP-1 and TIMP-1. Methods: A total of 30 healthy male rabbits were randomly divided into three groups: normal control group (group A), model group (group B), and Musk-Aconite-Earthworm pill group (group C) . KOA model was established by injecting papain to the right knees of rabbits. Rabbits in group C were intragastrically administered Musk-Aconlte-Earthworm pills for 4 weeks. Then, path-omorphological changes of cartilage and expression of MMP-1 and TIMP-1 were observed. Results: The degrees of pathological lesion in group C was less than that of group B (P〈0.05) . The expression of MMP-1 in group C was lower than that of group B (P〈0.01), and the expression of TIMP-1 in group C was higher than that of group B (P〈0.01) . Conclusion: Musk-Aconite-Earthworm pill can lessen cartilage cataplasia. The mechanism is possibly related to the regulation on MMP-1 and TIMP-1.
