摘  要:目的:探讨玉米须提取液对糖尿病大鼠肾脏的保护作用及机制研究。方法:采用健康雄性Sqrague-Dawley大鼠随机分为正常对照组、糖尿病组及玉米须治疗组,第4、8周任取6只大鼠,收集血、尿标本测定生化指标。大鼠肾组织分别作组织学、超微结构、免疫组织化学方法检测肾小球podocin蛋白表达,应用逆转录-聚合酶链反应(RT-PCR)观察podocin mRNA在肾组织中的表达。结果:在4周、8周,糖尿病组血糖、肌酐清除率、尿白蛋白排泄率较正常对照组均有不同程度的升高。玉米须提取液治疗组血糖、肌酐清除率、尿白蛋白排泄率较正常对照组均有不同程度的升高。玉米须提取液治疗组血糖、肌酐清除率、尿白蛋白排泄率显著低于同时期的糖尿病组。糖尿病组大鼠肾组织podocin mRNA和蛋白表达较正常对照组明显降低。玉米须提取液组明显上调。结论:玉米须提取液对糖尿病大鼠肾结构功能具有保护作用,可影响肾小球上皮细胞足突podocin蛋白表达,从而对糖尿病肾病有保护作用。Objective: To study the renal protective effect and the mechanism of Stigmata Maydis extract in STZ-induced diabetic rats. Methods: Healthy male Sprague-dawley rats were randomly divided into 3 groups: normal control group, diabetic group, and stigmata maydis group. At 4 and 8 week after the onset of diabetis, blood glucose (BG), screatinine clearance rate (Cer), 24h urinary albumin excretion rate (UAER) were measured. The renal cortical tissues were obtained and used for HE staining, immunohistochemical staining and electron microscopy. The expression of protein and mRNA of podoein in renal cortex were semi-quantitatively measured with RT-PCR. Results: At 2, 4 and 8 week, BG, Ccr and UKER were higher in diabetic group than in normal control group (P〈0.05). These parameters were significantly decreased in stigmats maydis group (P〈0.05) . RT-PCR analysis showed that the xpression of podocin mRNA in renal cortex was obviously decreased in diabetic group compared with that in normal control group. However, the expression of podocin mRNA increased in stigmata maydis group (P〈0.05) . Conclusion: Stigmata maydis has a protective effect on kidney of diabetic rats. The mechanism might be related to the inhibition of the expression of podocin mRNA in diabetic rats.
