摘 要:目的:观察哮喘时γ-干扰素(IFN-γ)和白细胞介素-4(IL-4)的关系及五味地龙汤对其影响。方法:取豚鼠48只随机分为正常对照组、哮喘模型组、地塞米松组、五味地龙汤高、中、低剂量组,每组8只。以卵白蛋白(OVA)腹腔注射致敏,气道吸入激发建立哮喘动物模型。致敏15d后,ig给药,连续8d。颈动脉取血离心取血清,用豚鼠IFN-γ和IL-4 ELISA试剂盒测定血清IFN-γ和IL-4水平。结果:五味地龙汤高剂量组豚鼠血清IFN-γ(35.06±2.83)pg.ml-1,中剂量组豚鼠血清IFN-γ(29.74±1.84)pg.ml-1,低剂量组豚鼠血清IFN-γ(28.80±1.36)pg.ml-1,地塞米松组血清IFN-γ(39.08±1.06)pg.ml-1,与正常对照组血清IFN-γ(35.41±2.31)pg.ml-1比较,无显著性差异(P>0.05);与哮喘模型组血清IFN-γ(18.74±2.22)pg.ml-1比较,均有非常显著的差异(P均<0.01)。五味地龙汤高剂量组血清IL-4(9.27±0.86)pg.ml-1,中剂量组血清IL-4(9.40±2.91)pg.ml-1,与正常对照组血清IL-4(9.45±0.62)pg.ml-1比较,无显著差异(P>0.05);与哮喘模型组血清IL-4(13.37±1.86)pg.ml-1比较,地龙汤高、中剂量组和地塞米松组均有非常显著的差异(P均<0.01);五味地龙汤低剂量组与哮喘模型组比较,亦有一定差异(P<0.05)。五味地龙汤高剂量组的作用优于低剂量组,组间比较,有一定差异(P<0.05)。结论:五味地龙汤能升高哮喘状态下豚鼠血清IFN-γ,降低IL-4水平,能调节哮喘状态下的IFN-γ/IL-4失衡。这可能是五味地龙汤抑制炎症介质的释放,治疗气道炎症的机制之一,其作用与剂量呈正比例关系。Objective: To explore the effect of Wuwei Earthworm decoction on serum interferon-γ (IFN-γ) and interleukine-4 (IL-4) at asthmatic attack. Methods: Forty-eight guinea pigs were randomly divided into six groups with 8 guinea pigs in each group: normal control group, asthmatic model group, dexamethasone group, Wuwei Earthworm decoction groups with low dose, middle dose and high dose. The asthmatic model of guinea pigs was established by intraperitionally injecting OVA and intranasally inhaling OVA. Fifteen days later, the guinea pigs were administered the decoction once a day for 8 days. The intracarotid blood was taken and centrifugated at 3000 γ min^-1 for 10 mins. Then, the levels of IFN-γ and IL-4 were determined with guinea pigs IFN-γ and IL-4 ELISA kits. Restdts: Levels of serum IFN-γ in earthwoml decoction group with high dose (35.06±2.83) pg·ml^-1, earthworm decoction group with middle dose (29.74± 1.84) pg·ml^-1, earthworm decoction group with low dose (28.80±1.36) pg·ml^-1, and dexamethasone group 139.08±1.06) pg·ml^-1 were significantly enhanced as compared with that in the model group ( 18.74±2.22) pg·ml^-1 (all P〈0.01) . But it was not significantly enhanced as compared with that in the normal control gronp (35.41±2.31 ) pg·ml^-1 (P〉0.05) . Levels of serum IL-4 in earthworm decoction group with high dose (9.27±0.86) pg·ml^-1, and earthworm decoction group with middle dose (9.40±2.91) pg·ml^-1 were significantly decreased as compared with that in the model group (13.37±1.86)pg·ml^-1 (all P〈0.01) . The effect in the high dose group was better than that in the low dose group. Conclusion: Wuwei Earthworm decoction can increase the serum IFN-γ level and reduce serum IL-4 level, and regulate the imbalance between IFN-γ and IL-4 at asthmatic attack.
