摘 要:目的探讨人尿道粘膜上皮细胞的体外培养技术与鉴定方法,为构建组织工程尿道提供种子细胞。方法取尿道下裂患者尿道粘膜,经消化离心后,接种于培养基中连续培养,观察细胞形态变化及生长、增殖过程,并绘制生长曲线;对细胞进行角蛋白免疫细胞化学染色和Giemsa染色。结果体外培养的细胞呈典型的铺路石样外观,可传至8代;角蛋白免疫细胞化学染色呈阳性;8代细胞为正常2倍体核型。结论人尿道粘膜上皮细胞可在体外进行连续培养,本实验培养的细胞可用于构建组织工程尿道。Objective To investigate a method of culturing and identifying human urethral epithelium, provide seed cells for tissue engineering urethra. Methods Specimens from urethral epithelium of hypospadias were isolated, digested and cultured. We observed the cell morphology, growth and proliferation, and protracted growth curve. The cells were stained by immunocytochemistry and Giemsa. Results The cultured urethral epithelium cells could be subcultured for 8 generations, with the same size as road rocks. The cells showed positive staining for cytokeratin, and they maintained diploid cells at 8 passages. Conclutions The culture technique for human urethral epithelium has been established successfulky, the culturing cells could be used in construction of tissue engineering urethra.
