摘  要:目的探讨一步法逆转录聚合酶链反应(one-step reverse transcriptase-PCR或一步法RT-PCR)在人狂犬病检测中的应用。方法4例临床诊断狂犬病死亡患者的冰冻脑组织,用一步法RT-PCR检测病毒,狂犬病毒CVS株感染的鼠脑和急性胰腺炎死者的脑分别作为阳性和阴性对照。结果4例脑组织中提取的RNA,电泳后得到与狂犬病N基因长度一致的片断。结论一步法RT-PCR可用于人狂犬病毒的检测,在法医学鉴定中具有重要意义。Objective To discuss the application PCR) in the detection of human rabies. Methods The of one-step RT-PCR(one-step reverse transcriptasefreezing identified by clinical diagnosis were collected and were detected brain tissues of four patients died of rabies by one-step RT-PCR. The rat brain infected by CVS strain of rabies virus was used as a positive control. The brain of a patient died of acute pancreatitis was used as a negative control. Results The RNA could be extracted from the freezing tissues. After one-step RT-PCR, specific bands with the expected size of the N gene could be detected in the samples from all the 4 cases. Conclusion One-step RT-PCR could be used in detection of human rabies. It was important for forensic identification.
