摘  要:RNA干扰(RNA interfering,RNAi)是由双链RNA介导的序列特异性转录后的基因沉默现象,是生物界一种古老而进化上高度保守的基因表达调节机制。随着分子生物学理论与技术研究的不断深入,近几年利用RNAi技术特异性剔除或关闭特定基因表达而防治疾病成为医学界研究的热点。信号转导及转录激活因子1(signal transduction and activator of transcription-1,STAT1)是一种能被多种配体激活而使相关免疫应答基因表达的转录因子。STAT1的激活可诱发支气管哮喘气道的炎症和气道高反应性。利用RNAi技术沉默STAT1基因表达,阻止气道变应性炎症反应,对防治支气管哮喘的发生发展具有重要的临床意义。RNA interference(RNAi) is a gene silencing phenomenon,which happens after sequence- specific transcription mediated by double-stranede RNA. And it is an ancient and high conserved on evolution mechanism of regulating gene expression in living nature. With the development of the theory and technology of molecular biology,it becomes research focus in medical domain that to prevent and treat diseases by the way of specificly rejecting or shuting off specified gene expression using the technology of RNAi in recent years. Signal transducer and activator of transcription-1 (STAT-1) is a transcription factor, which can be activated by a variety of ligands and result in the expression of relative immune response genes. The activation of STAT-1 can induce the inflammation of bronchial asthma and airway hyperreactivity. It has important clinical significance in preventing and curing bronchial asthma that to make the expression of STAT-1 gene silence with the technology of RNAi so as to interrupt the airway allergy inflammation.
