摘  要:目的探讨鳖甲煎丸对人肾小球系膜细胞(HMC)基质金属蛋白酶系统的影响。方法应用细胞培养技术,进行HMC培养,采取血清药理学方法,制备鳖甲煎丸药物血清,利用脂多糖(LPS)为刺激因子,应用酶联免疫吸附试验(ELISA)法检测基质金属蛋白酶-9(MMP-9)及其抑制物金属蛋白酶组织抑制因子-1(TIMP-1)的表达。结果各组MMP-9的含量比较无明显差异(P>0.05);LPS组TIMP-1的含量与空白组比较明显升高(P<0.01);鳖甲煎丸组与空白组比较,TIMP-1含量明显降低(P<0.05);脂甲组、脂坦组与LPS组比较,TIMP-1表达明显降低(P<0.01)。结论LPS具有促进TIMP-1表达的作用,对MCsMMP-9作用较小。鳖甲煎丸及缬沙坦药物血清具有抑制正常培养条件及LPS刺激条件下TIMP-1表达的作用,这可能是其延缓肾小球硬化的部分作用机制。Objective To investigate the influence of Biejiajian Pills (BP) on matrix metalloproteinase system of human mesangial cells (HMC). Methods HMC were cultured by applying cell cultural technique. The serum pharmacological method was used in the preparation of BP serum. The expressions of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) were detected by enzyme-linked immunosorbent assay (ELISA) taken lipopolysaccharide (LPS) as stimulating factor. Results The contents of MMP-9 in every group were not different ( P 〉 0.05 ). The content of TIMP-1 in LPS group were obviously higher than that in the blank group ( P 〈 0.01 ) , that in BP group was obviously lower than that in the blank group ( P 〈 0.05 ). The expression of TIMP-1 in the LPS combined BP group or in the valsartan combined BP group was obviously lower than that in the LPS group (P 〈 0.01 ). Conclusion LPS has an improving effect on the expression of TIMP-1, but less effect on the expression of MMP-9. The serum of BP or serum of valsartan can inhibit the expression of TIMP-1 in the condition of normal or LPS stimulation, which may be the mechanism of BP in the delay of glomerular sclerosis.
