摘 要:目的研究小檗碱(Ber)对血管紧张素Ⅱ(AngⅡ)诱导的心肌成纤维细胞(CFB)增殖和胶原合成的影响,并探讨其作用机制。方法采用酶消化法和差速贴壁法获得纯化的CFB,应用MTT法测定CFB增殖、羟脯氨酸法测定胶原蛋白含量、化学比色法测定NO含量和NOS活力、ELISA法测定TGF-β1含量。结果小檗碱在1.25-10 mg/L浓度范围内可明显抑制AngⅡ诱导的CFB增殖和胶原蛋白合成增加,同时可升高NO含量和NOS活力、降低TGF-β1含量。结论小檗碱可抑制AngⅡ诱导的心肌成纤维细胞增殖和胶原蛋白合成的作用,对抑制心肌纤维化,改善心室重构有积极意义,其作用机制与促进心肌成纤维细胞分泌NO、减少TGF-β1含量有关。Objective To study the influence of berberine (Ber) on cardiac fibroblasts (CFB) proliferation and collagen protein synthesis induced by Angiotensin Ⅱ , and investigate its mechanism. Methods Purified CFB were obtained by the methods of digestive enzyme and differential adhesion. CFB proliferation was determined using MTr method. Collagen content was determined using hydroxproline method. NO content and NOS activity was determined using chemistry colorimetry method. TGF-β1 content was determined using enzyme linked immunosorbent assay (ELISA). Result Berberine at the concentration of 1.25 - 10 mg/L could obviously inhibit CFB proliferation and the increased collagen protein synthesis induced by Ang Ⅱ , and also could increase NO content and NOS activity and decrease TGF-β1 content. Conclusion Berberine has inhibitiory effects on CFB proliferation and collagen protein synthesis induced by Ang Ⅱ , which is of positive significant to myocardial fibrosis inhibiting and ventricular remodeling improving . The mechanism might be related with increasing NO secretion and reducing TGF-Ⅱ expression of CFB.
