摘 要:背景:前期实验已成功制备纳米抑菌复合材料——丁胺卡那-蒙脱土复合材料。目的:在前期实验基础上制备丁胺卡那-蒙脱土/硅橡胶片抗菌复合导尿管材料,测试其相关热学、力学性能以及抗菌性能,并与全硅橡胶材料相比较。设计、时间及地点:对比分析,体外实验,于2006-05/2007-04在香雪制药股份有限公司实验室完成。材料:自制丁胺卡那/蒙脱土复合材料;甲基乙烯基硅橡胶为江西宏达化工新材料股份有限公司生产。方法:取生胶和抗菌负载蒙脱土用双辊开炼机混炼,然后加入硫化剂,用平板压力机压板制样,硫化温度170℃,硫化时间10min,制得4个不同胶料配方的胶,分别为每100份硅橡胶中添加0,5,7.5,10份丁胺卡那/蒙脱土复合材料。主要观察指标:差示扫描量热仪表征其热学性能,偏光显微镜观察微观结构;通过体外释放及抑菌实验检测抗菌活性和缓释效果。结果:丁胺卡那-蒙脱土复合材料/硅橡胶结晶温度不同程度向低温迁移,其中每100份硅橡胶中添加7.5份丁胺卡那/蒙脱土复合材料变化较大,达到-38.50℃,材料的力学性能有不同程度的提高,有效抗菌时间可以达到21d。结论:丁胺卡那-蒙脱土复合材料/硅橡胶导尿管材料具有良好的力学性能、抗菌性能及缓释效果。BACKGROUND: Previously, amikacin/organo-montmorillonite (OMMT) has been successfully prepared as an antibacterial nano-composite material. OBJECTIVE: To prepare silicon rubber/amikacin-OMMT composite as an antimicrobial composite catheter, to test its properties of thermology, mechanics and antibiosis, and to compare it with pure silicon rubber. DESIGN, TIME AND SETTING: Contrast analysis experiments in vitro were carried out in the laboratory of Xiangxue Pharmaceutical Stock Co.,Ltd from May 2006 to April 2007. MATERIALS: Methyl vinyl silicone rubber was provided by Jiangxi Hongda New Chemical Materials Co., Ltd. Amikacin-OMMT composite was self-made. METHODS: Raw silicon rubber and OMMT composites were mixed by a double-roller open refiner, and then sulfurized with the added vulcanizer at 170 ℃for 10 minutes. Four kinds of film samples with different adhesive masses were prepared by a flat plate piercer, every 100 samples of silicon rubbers were seperatively supplemented with 0, 5, 7.5, 10 samples of amikacin-OMMT composite. MAIN OUTCOME MEASURES: Differential scanning calorimeter was used to detect thermotical property; polarizing microscope was utilized for the microstructure observation; Antimicrobial activity and slow-release effect were determined by in vitro bacteriostasis and release, respectively. RESULTS: The crystallization temperature of silicone rubber/amikacin-OMMT composite decreased at different degrees, with the lowest temperature -38.50℃ at 7.5 phr. The mechanical property of the synthetical material was improved, and the antibiosis activity lasted for 21 days at the most. CONCLUSION: The silicone rubber/amikacin-OMMT composite shows that its mechanical property, antimicrobial activity and slow-release effect are satisfying.
