摘  要:目的研究糖尿病大鼠心肌胆固醇转出相关基因表达的变化。方法用STZ选择性破坏大鼠胰岛β细胞,建立实验性糖尿病大鼠模型,动态观察分析糖尿病不同病程中大鼠心肌胆固醇转出相关基因过氧化物酶体增殖物活化受体α(PPARα),肝X受体α(LXRα),ATP结合盒转动体A1(ABCA1)mRNA表达以及胆固醇含量的变化。结果糖尿病12~17w时大鼠心肌PPARα激活,LXRα、ABCA1 mRNA表达增加,但心肌胆固醇含量没有变化。结论STZ诱导的糖尿病大鼠心肌随着PPARα的激活,LXRα、ABCA1 mRNA表达增加,PPARα、LXRα共同参与了胆固醇代谢。Objective To investigate changes in mRNA expression of cardiac genes involved in cholesterol efflux in STZ-induced diabetes in rats. Methods Experimental diabetes model was induced by STZ destroying insular β-cell selectively in rats. Cholesterol content of myocardial tissue was measured by auto-biochemical-analyst using enzymatic procedure and the mRNA levels of PPARα, LXRα and AB- CA1 in myocardial tissue were detected by RT-PCR during course of diabetes. Results PPARα was activated and the levels of LXRα, AB- CA1 mRNA increased at the 12th and 17th week, but no changes in cholesterol content were found in myocardial tissue of diabetic rats. Conclusions mRNA expression of LXRα and ABCA1 are increased along with the activation of PPARα in myocardium of diabetic rats. PPARα and LXRα together participate in cholesterol metabolism.
