摘 要:分别用二乙烯三胺、丙醛对壳聚糖的氨基进行改性,并与NO气体在甲醇钠/甲醇溶液中6×101.3kPa反应7 d,得到两种改性壳聚糖/一氧化氮加成产物。采用FT-IR、UV1、H-NMR和13C-NMR对产物的化学结构进行了表征,并用Griess试剂法测定了其NO释放曲线。NO释放试验表明,增加改性壳聚糖亲核位点NH的数量可以显著提高NO负载量。CS/DETA/NO的NO最大释放量为1945 nmol/mg,CS/PA/NO的NO最大释放量是1156 nmol/mg。diethylenetriamine (DETA) and propionic aldehyde (PA) were employed individually to modify the primary amine groups on chitosan. Two diazeniumdiolates (ehitosan/NO adducts) were prepared by suspending the chitosan derivatives in sodium methoxide-methanol solution at 6 atm for seven days, and their chemical structures were identified by FT-IR, UV and ^1H-NMR and ^13C-NMR. The NO release profiles for ehitosans derivatives in PBS solution (pH 7.4, 37 ℃ ) were investigated by the Griess assay. The results show that the increase of nucleophilic sites on DETA modified chitosan results in high loadings of NO, the maximum NO release of CS/DETA/NO and CS/PA/NO is 1945 nmol/mg and 1156 nmol/mg respectively.
