摘 要:对传统药食两用植物落葵的水溶性多糖提取工艺进行研究。采用热水浸提、石油醚脱脂、Sevage法脱蛋白、乙醇沉淀等方法来提取落葵粗多糖。采用单因素试验和正交试验L9(34)研究料液比、温度、时间对多糖提取率的影响,以苯酚-硫酸显色法测定多糖浓度。结果表明,料液比和提取温度为影响落葵多糖提取的主要因素。正交设计得到最佳提取工艺为:料液比1∶50,提取时间5h,提取温度85℃。多糖提取率最大为4.0%;超声波提取在温度70℃,超声功率160W,时间120min时可达最大,其提取率为17.02%,是热水浸提法的4.25倍,这表明超声提取法是一种更为有效的落葵粗多糖提取方法。To study the extraction technology of water-soluble polysaccharide from Basella.rubra, which is a kind of Chinese traditional medicine. The water-soluble polysaccharide was extracted by hot water, then removed the protein by sevag method, and fattiness by petroleum ether, and precipitated with ethanol. Single factor test and orthogonal experiment design methods L9 (34)were applied to analyze the influence of the factors such as solid-liquid ratio ,the extraction temperature and the extraction time on the extraction yield of the polysaccharides. The ultrasonic extraction of water-soluble polysaccharide was studied under set conditions.The content of polysaccharide was determined by phenol-sulfuric acid colormetry. The experimental results indicated that the extraction time and the extraction temperature would significantly affect the extraction percent of the polysaccharides.The optimum solid-liquid ratio was 1:50, the extraction time was 5 h and the extraction temperature was 85 ℃. The water-soluble polysaccharide extraction yield is 4.0 %; under the time of 120 min and power of 160 W, the ultrasonic extraction can obtain higher polysaccharide extraction yield with 17.02 %, nearly 4.25 times as much as that from hot water extraction,which indicates that the ultrasonic extraction is a better method for polysaccharide extraction of Basella.rubra. The extraction technology of water-soluble polysaecharide from Basella.rubra supplies the bases for the active research of Basella.rubra.
