摘 要:为探讨中缝大核(nucleusraphesmagnus,RM)内生长抑素(sonatostatin,SOM)在针刺镇痛中的作用,本文用原位杂交组织化学方法(ISHH)和免疫细胞化学(IC)相结合的双重标记技术.观察了大鼠RM内SOMmRNA在正常、疼痛刺激、单纯电针刺激和电针镇痛下的变化特点。结果发现:典型SOMmRNA阳性细胞胞体及突起近端内合蓝紫色颗粒.胞核不着色呈空泡状。正常组RM内SOMmRNA阳性细胞数量少,形态多样,但多为校形、三角形,多分布于RM的双翼。大多数细胞显示出1~3个突起,细胞染色较淡。和正常组相比,疼痛组和单纯电针组RM内SOMmRNA阳性细胞明显增多,染色加深。经图像分析和统计学处理,阳性细胞个数和平均光密度变化均P<0.01。在电针镇痛下,RM内SOMmRNA阳性细胞显得更为密集,且测得的阳性细胞个数和平均光密度与其他三组相比,在统计学上均有显著性差异(P<0.01)。在正常情况下,RM内有少量的SOMmRNA与5—HT双标记细胞,在棕黄色的胞浆背底中有蓝紫色颗粒。与正常组相比,疼痛刺激组和单纯电针组双标细胞增多(P<0.01),电针镇痛后更甚,与其他三组相比P<0.01。以上结果提示:1.SOM可能参与痛觉调制。2.SOM可能参与电针镇痛。3.RM内SOMmRNA与5-HT共存,二者可能协同作用,共同参与电针镇痛?By using the method which combines in situ hybridization with immunohistochemistry ABC, theexpression of somatostation (SOM) mRNA and its coexistence with 5-HT in magnus raphes raphes nucleus (RM) of therat were observed in the normal group, pain group, simple electroacupuncture group and electroacupuncture analgesia group (EA). Fusiform and trianguular cells with a weak hybridization signal could be onserved and distributed chiefly in lateral part of RM. An increase of SOM mRNA positive cells could be seen in pain group and simpleelectroacupuncture group as compared with the control group, and was statistically significant. In EA group thenumbers of SOM mRNA positive cells increased more than any other groups (P< 0. 01 ). A few SOM mRNA and5-HT double labelled cells could be seen in RM in control group. Most of the double labelled cells were distributed inin lateral Part Of RM. Compared with control group, double labelled cells increased in pain group (P< 0. 01 ) andsimple electroacupuncture group (P< 0. 01 ) and increasd significantly in EA group (P < 0.01). The resultS suggest: 1. SOM takes part in pain modulation. 2. SOM plays a role in EA. 3. Coexistence of SOM mRNA and 5 HT in RM suggests that SOM and 5-HT may have some bearing on their synergistic action in EA.
