摘  要:目的探讨红参水提物对Aβ25-35诱导人神经瘤母细胞SH-SY5Y细胞凋亡的保护作用。方法用Aβ25-35处理SH-SY5Y细胞,模拟阿尔茨海默病患者脑内神经元的病理损伤模型,以不同浓度的红参水提物进行干预;用倒置显微镜观察其形态学的变化;MTT法测定细胞的存活率;流式细胞技术检测细胞的凋亡率以及线粒体膜电位的变化。结果50μmol·L-1Aβ25-35诱导SH-SY5Y细胞72h后,细胞变圆、聚集,其存活率为39.26%±3.16%、凋亡率为37.30%±0.69%,线粒体红绿荧光强度比值为0.45±0.10;而Aβ25-35诱导的SH-SY5Y细胞与不同浓度(1、5、10mg·ml-1)红参水提物同时孵育后,明显减少了细胞损伤,升高了细胞存活率、降低了凋亡率,并升高了线粒体红绿荧光强度比值。结论红参水提物对Aβ25-35诱导的SH-SY5Y细胞凋亡有显著的保护作用。OBJECTIVE To study the protective mechanism of water extracts of Red Ginseng on Aβ25-35 - induced human neuro- blastoma SH - SYSY cells apoptosis. METHODS SH - SYSY cells were treated with Aβ25-35 . We simulated the pathologic damage in brain of patients with Alzheimer' s disease, and different doses of water extracts of Red Ginseng were added. The morphologic changes were observed by inverted microscope. Cell survival rate was measured by MTr assay. The percentage of apoptosis and the changes of mitochondrial membrane potential were measured by flow cytometry. RESULTS After 72 h of Aβ25-35 (50 μmol. L^-1 ) treatment, SH - SYSY ceils became rounder and aggregated. The cell survival rate was 39.26% ± 3. 16% ; the percentage of apoptosis was 37.30% ±0.69%, and the mitochondrial membrane potential was 0.45 ± 0.10. When incubated with Aβ25-35with different doses of Red Ginseng extract ( 1,5,10 mg.ml^-1 ) for 72 h, cell damage was obviously decreased. Cells survival rate increased ; cell apoptosis decreased, and mitochondrial membrane potential increased as well. CONCLUSION Water extracts of Red Ginseng has remarkable neuroprotective effects on SH - SY5Y cells apoptosis induced by Aβ25-35.
