摘  要:目的从分子水平上阐释损伤血淤证的本质;揭示桃红四物汤治疗损伤血淤证的机制。方法用定量化重力击打法制备损伤血淤证模型,造模后按要求灌胃,收集血液,检测血浆中TXB2、6-keto-PGF1α的含量。结果模型组在2、8、24h时,大鼠血浆中TXB2的含量先升后降,与正常对照组比较P<0.05;与模型组同时间点比较,治疗组2、8、24h的大鼠血浆中TXB2的含量升高(P<0.01)。与正常对照组比较,模型组2h时的6-keto-PGF1α含量显著降低(P<0.01);与模型组同时间点比较,治疗组48h时的6-keto-PGF1α含量降低(P<0.05),168h的6-keto-PGF1α含量升高(P<0.05)。结论损伤血淤证模型大鼠血浆中TXB2、6-keto-PGF1α的含量呈现不同的时相性变化;桃红四物汤对损伤血淤证的模型大鼠血浆中TXB2、6-keto-PGF1α均具有调节作用,但其作用时相不同。OBJECTIVE To explore the essential of traumatic injury of blood stasis syndrome from the aspect of molecule and the molecular mechanism of Taohong Siwu decoction (THSWD). METHODS Eighty - eight Sprague - Dawley (SD) rats were randomly divided into control, model and THSWD groups at 2,8,24,48 h, 168 h. The model and THSWD groups were made of closed haematoma by quantitative beating, then, the rats of THSWD groups were fed according to the request and the rats' blood was collected after 2,8, 24,48,168 h and they were detected by radio immunology. The results were analyzed by SPSS13.0. RESULTS Result of TXB2 :Compared with normal group,the expression of TXB2 in serum increased at first, then decreased at 2,8,24 h after injury(P 〈0. 05 ). Compared with model group at the same time point, the expression of TXB2 in serum of THSWD group at 2,8,24 h after injury was extremely higher ( P 〈 O. 01 ). Result of 6 - keto - PGF1α: compared with the normal group, at first, expression of 6 - keto - PGF1α in plasma of model group at 2 h after injury extremely decreased ( P 〈 0. 01 ). Compared with model group at the same time point, the expression of 6 - keto - PGF1α in plasma of THSWD group at 48 h was drastically lower( P 〈 0. 05 ), the expression of 6 - keto - PGF1α in serum of THSWD group at 168 h after injury was higher( P 〈 0. 05 ). CONCLUSION The expressions of TXB2,6 - keto - PGFI, in serum of animal model of traumatic injury with blood stasis syndrome were different time - linked. THSWD could regulate the expressions of TXB2 ,6 - keto - PGF1α in plasma of the model at different time course.
