摘  要:目的探讨三氧化二砷(ATO)对MRL/lpr自发性狼疮小鼠脾脏单个核细胞(SMC)中DNA甲基转移酶1(DNMT1)和黏附分子淋巴细胞功能相关抗原-1(LFA-1)的亚基CD11a表达的影响及其可能作用机制。方法分离小鼠SMC,采用20g/ml植物血凝素和1000IU/ml白介素-2体外诱导培养SMC 48h后,随机分为空白组、ATO组和5-氮胞苷(5-Az)组,继续培养24h,采用RT-PCR法检测两组小鼠SMC中DNMT1及CD11a基因转录水平的表达情况。结果MRL/lpr小鼠ATO组中DNMT1-mRNA的灰度比值较5-Az组增高(P<0.05),MRL/lpr小鼠ATO组中CD11a-mRNA的灰度比值较5-Az组降低(P<0.05)。结论ATO治疗系统性红斑狼疮的作用机制可能与其能逆转低甲基化状态有关。Objective To investigate the effect of arsenic trioxide (ATO) on the expression of DNA methyltransferasel (DNMT1)mRNA and CD11a mRNA in splenic mononuclear cells (SMNC)of spontaneous lupus nephritis MRl_/Ipr mice. Methods Isolated SMNC of MRL/Ipr and C57BL mice were induced by PHA 20g/ml + IL-2 10001U/rot for 48 h, and the cultured cells were randomly divided into control group, ATO group, 5-AZ group. Each group was incubated in vitro for 24h and RNA was extracted. The expression of DNMT1 and CD11a was determined by RT-PCR. Results The gray scale value of DNMTI-mRNA in ATO group was significantly higher than that in 5-Az group (P 〈 0.05)and the gray scale value of CD11a-mRNA in ATO group was significantly lower than that in 5-Az group ( P〈 0.05). Conclusion The therapeutic effect of ATO for SLE may be associated with reversed low DNA methylation.
