摘  要:目的探讨参附注射液对失血性休克大鼠血清和人脐静脉内皮细胞(HUVEC)中血栓调节蛋白(TM)、内皮细胞蛋白C受体(EPCR)表达的影响。方法将50只SD大鼠随机分为正常对照组、假休克组、失血性休克组、林格注射液治疗组(林格组)、林格注射液加参附注射液治疗组(参附组),分别检测各组可溶性TM(sTM)、可溶性EPCR(sEPCR)水平及TM、EPCR mRNA和蛋白表达水平。结果与假休克组比较,休克组和林格组sTM、sEPCR及TM、EPCR mRNA含量均增高(P<0.05),TM和EPCR蛋白含量均降低(P<0.05)。与林格组比较,参附组sTM、sEPCR含量和TM、EPCR mRNA含量均下降(P<0.05),TM和EPCR蛋白含量均升高(P<0.05)。结论参附注射液可从基因、蛋白水平影响人血清和HUVEC中TM和EPCR的表达,从而阻止失血性休克的进展。Objective To investigate the effects of Shenfu injection on thrombomodul (TM) and endothelial cell protein C receptor (EPCR) levels in serum of hemorrhagic shock rats and human umbilical vein endothelial cells. Methods Fifty rats were randomly divided into five groups: normal control group, sham shock group, hemorrhagic shock group, Linger-treated group and Shenfu-treated group. The soluble thrombomodulin (sTM) , endothelial protein C receptor ( sEPCR),TM, EPCRmRNA and protein were detected by enzyme linked lmmunosorbent assay (ELISA), reverse transcription polymerase chain reaction (RT-PCR), western blotting (WB), respectively. Results The mean levels of sTM, sEPCR and the expression of TM, EPCR mRNA in hemorrhagic shock and Linger-treated group was significantly higher than that in sham shock ( P〈 0.05),while the level of protein was lower than that in sham shock (P〈0.05). The mean levels of sTM, sEPCR and expression of TM, EPCR mRNA in Shenfu-treated group was significantly lower than that in Linger-treated group ( P〈 0.05),while the level of protein was higher than that in Linger-treated group (P〈0.05). Conclusion Results suggest that Shenfu injection can influence TM and EPCR mRNA and protein expression, therefore it might be effective in prevention of development of hemorrhagic shock.
