摘  要:目的:探讨人参皂甙Rb1对老年性痴呆模型大鼠顶叶皮质β-分泌酶及早老蛋白-1(PS-1)表达水平的影响。方法:30只SD大鼠随机分为对照组、模型组和治疗组。模型组大鼠胃饲氯化铝200mg·kg-1.d-1,同时腹腔注射D-半乳糖60mg·kg-1.d-1;治疗组大鼠于造模结束后,腹腔注射人参皂甙Rb110μg·kg-1.d-1。采用免疫组织化学方法和图像分析技术检测β-分泌酶及PS-1的表达,通过Morris水迷宫测试观察大鼠学习记忆能力的改变。结果:与对照组比较,模型组β-分泌酶及PS-1的表达明显增多,学习记忆能力明显下降;与模型组相比,治疗组β-分泌酶及PS-1的表达明显减少,学习记忆能力在第2天及第3天显著提高。结论:人参皂甙Rb1可以抑制β-分泌酶及PS-1的表达,对老年性痴呆模型大鼠有一定的防治作用。Objective: To investigate the effect of ginsenoside Rb1 on the expression of β-seeretase and presenilin-1 (PS-1) in the parietal lobe of Alzheimer disease-like rats. Methods: Thirty SD rats were randomly divided into a control group, a model group and a treatment group. The model rats were established with administration of I)-galactose 60 mg· kg-1· d- 1 and aluminum chloride 200 mg·kg -1 · d-1 for 2 months, the control group were given the same dose of sodium chloride, and treatment group were given ginsenoside Rb1 10μg · kg-1· d -1 for 4 weeks after the model rats were established. The expressions of β-seeretase and PS-1 in the parietal lobe were detected by immunohistochemistry with image analysis. Morris test was used to determine the ability of learning and memory. Results: Compared with the control group, the levels of β-secretase and PS-1 were significantly higher in the model group (P〈0.05), and learning and recognition ability in this group was obviously worse (P〈0.05) ; Compared with the moctel group, the levels of β-secretase and PS-1 were significantly low- er in the treatment group (P〈0.05), and learning and recognition ability in this group was better. The learning and recog- nition ability was obviously improved on the second and the third day (P〈0.05). Conclusion: Ginsenoside Rbl has preven- tive and therapeutic effect on Alzheimer disease-like rats, via inhibiting the expression of β-seeretase and PS-1.
