摘 要:目的:探讨创伤弧菌(Vibrio vulnificus,VV)脓毒症大鼠肺组织脂多糖受体CD14 mRNA、核因子-κB(NF-κB)基因的表达及其动态变化。方法:SD雄性大鼠60只,随机分为6组,每组10只。第1组:正常对照组(NC组);第2~6组为VV组:予大鼠下肢皮下注射VV,造成VV脓毒症模型,以注射VV后2、6、9、12、16h各时间点分别为2~6组。取各组大鼠部分肺组织,用RT-PCR法测定大鼠肺组织不同时间点CD14 mRNA、NF-κB mRNA的表达。结果:NC组大鼠肺组织有少量CD14、NF-κB mRNA表达,分别为0.392±0.056和0.366±0.179。感染VV后大鼠肺组织CD14 mRNA表达增加,2、6、9、12、16h的表达量分别为0.976±0.180,1.148±0.218,1.026±0.033,0.988±0.129,0.970±0.125。大鼠肺组织NF-κBmRNA表达量也明显增加,2、6、9、12、16h的表达量分别为0.763±0.215,1.100±0.101,1.068±0.174,1.057±0.076和0.800±0.205。与NC组相比,二者均明显升高(P<0.05)。结论:VV脓毒症大鼠肺组织CD14、NF-κB的基因表达水平在脓毒症中明显升高,其与脓毒症的发生、发展密切相关,动态监测其改变对VV脓毒症发病过程具有一定的预警意义。Objective: To detect the Gene expression and the dynamic change of pulmonary li-popolysaccharide receptor CD14 and NF-KB in rats with Vibrio vulnificus (VV) sepsis. Methods: Sixty male Sprague-Dawley (SD) rats were equally divided into six groups randomly. The first group was named control group. W groups include the second to sixth group which were injected W to make W sepsis model. Samples were taken from control group and the second group to the sixth group at the timepoint of 2, 6, 9, 12, 16 h after injection respectively, the genes of pulmonary lipopelysaccharide receptor CD14 and NF-κB in rats lung were measured in the different time by the method of RT-PCR. Results: The Gene expression of pulmonary lipopolysaccharide receptor CDI4 and NF-κB in the control group was a low level (0.392±0.056, 0.366±0.179), but they increased significantly at 2 h (0.976±0.180, 0.?63±0.215), 6 h (1.148±0.218, 1.100±0.101), 9 h (1.026±0.033, 1.068±0.174), 12 h (0.988±0.129, 1.057±0.076), 16 h (0.970±0.125, 0.800±0.205) after VV being injected (P〈0.05). Conclusion: Our data suggest that the Gene expression of pulmonary lipopelysaccharide receptor CD14 and NF-κB elevation are associated with the development of VV sepsis. Monitoring the change of the Gene expression of pulmonary lipepolysaccharide receptor CD14 and NF-κB in the lung have the benefit of mastering the VV sepsis progress.
