摘 要:目的研究红细胞表面跨膜糖蛋白标准型CD44(CD44s)对红细胞黏附胃癌细胞的影响。方法利用流式细胞分析法测定正常人红细胞表面CD44s表达情况;利用免疫组化方法检测胃癌细胞表面透明质酸(HA)表达情况;以胃癌细胞红细胞花环形成率来评价红细胞对胃癌细胞的黏附能力。结果正常人红细胞表面CD44s平均荧光强度为308.694±8.643;胃癌细胞表面的透明质酸平均IOD值为4617±1775.363;经鼠抗人CD44s单克隆抗体处理过的正常人红细胞黏附胃癌细胞的能力显著下降,胃癌细胞红细胞花环形成率为(20.6±9.232)%,明显低于空白对照组(P<0.05)。结论红细胞表面CD44s在红细胞免疫黏附胃癌细胞的过程中发挥一定的作用。Objective To study the effect of standard isofonn of CD44 (CD44s) in erythrocyte surface on the immune adherence of erythrocytes to tumor ceils. Methods The expression of CD44s in the plasma membrane of normocyte was detected by using flow cytometry; The expression of HA in the plasma membrane of gastric carcinoma cell was detected by immunohistochemical staining;The immune adher- ent ability of normocytes to gastric carcinoma cells was detected by using rosette rate of normocytes immune-adhering tumor cells (R-T rosette ). Results The mean fluorecence intensity of CD44s in the normocytes plasma membrane was (308.694±8.643).The mean IOD of HA in the gastric carcinoma cells was (4617±1775.363)o The ability of nomlocyte immune-adhering gastric carcinoma cell was significantly lower after normocytes were treated by mouse anti-human CD44s monoclonal antibody.The R-T rosette was 20.6±9.232 (%),which was less than the blank control group (P〈 0.05). Conclusion CD44s in the erythrocyte may play an important role on the immune adherence of eryhrocytes to tumor cells.
