摘 要:目的观察γ-羟丁酸钠对大鼠肝脏缺血再灌注损伤中的保护作用及其可能机制。方法36只雄性Wist-ar大鼠随机分成3组,每组12只。假手术组:开腹显露第一肝门后关腹;缺血再灌注组:开腹显露第一肝门后,用无损伤血管钳夹闭肝蒂,阻断血流30 min后,放开血管钳再灌注2 h,制作常温下大鼠肝脏热缺血模型;γ-羟丁酸钠处理组:麻醉后经静脉泵注γ-羟丁酸钠60 mg/kg,余同缺血再灌注组。分别于缺血再灌注6 h后抽血检测血清AST、ALT、SOD及MDA水平,并取肝组织行病理组织学检测和Westernblot法检测HMGB-1水平。结果缺血再灌注组AST、ALT、MDA及HMGB-1的含量均高于假手术组和γ-羟丁酸钠组(P<0.01)。而γ-羟丁酸钠组SOD的活性高于缺血再灌注组(P<0.05)。结论γ-羟丁酸钠预处理对肝脏缺血再灌注损伤具有明显的保护作用。Objective To study the protective effects of sodium hydrobutyrate on liver ischemic reperfusion injury and its mechanism in rats. Methods 36 healthy male Wistar rats were randomized into three groups: Control group (C group), ischemic reperfusion group (IR group) and ischemic hydrobutyrate preconditioning group (P group), each consisting of 12 animals. IR group and P group were made liver ischemic reperfusion injury model, and P group were pretreated with sodium hydrobutyrate 60 mg/kg, The plasma ALT, AST, SOD and MDA levels were measured, and HMGB -lin the liver tissue were also observed using western blot methods. Results The plasma ALT,AST and MDA levels in IR group were significantly higher than those in C group and P group(P 〈0.01) ,and SOD levels in IR group was higher than that in C group and P group(P 〈0.05). HMGB - 1 had no expression in C group, but its mRNA was higher in IR group than in P group(P 〈0.01 ). Conclusion With the sodium hydrobutyrate pretreatment, the liver tissue was protected efficiently against ischemic reperfusion injury.
